Recent progress in NIR-II fluorescence imaging-guided drug delivery for cancer theranostics

Summary: Fluorescence imaging in the second near-infrared window (NIR-II) has many advantages, making it a popular choice for imaging-guided drug delivery (IGDD) paradigms. This review discusses the recent progress in NIR-II IGDD-mediated cancer therapeutics, including design approaches, different therapies, and updated platforms. The review also highlights the innovation directions for NIR-II IGDD platforms to improve theranostic outcomes and clinical translation. This review will contribute to the development of safe and efficient NIR-II fluorescence-mediated anticancer drug delivery.

Doubly Strapped Zwitterionic NIR-I and NIR-II Heptamethine Cyanine Dyes for Bioconjugation and Fluorescence Imaging

Summary: This study reports a synthetic method for producing novel doubly strapped zwitterionic heptamethine cyanine dyes with enhanced stability. These dyes, including a structural analogue of ZW800-1, can be conjugated to proteins, including monoclonal antibodies, without causing fluorophore degradation or dye stacking. The fluorescent antibody conjugates show excellent tumor-targeting specificity in a xenograft mouse tumor model. The enhanced stability provided by the doubly strapped molecular design enables new classes of in vivo NIR fluorescence imaging experiments with possible translation to humans.

Highly Twisted Conformation Thiopyrylium Photosensitizers for In Vivo Near Infrared-II Imaging and Rapid Inactivation of Coronavirus

Summary: We developed efficient heavy-atom-free photosensitizers in the near-infrared (NIR) range using a facile approach. By modifying the thiopyrylium-based NIR-II dyads, our study showed that a sterically bulky and electron-rich moiety enhanced the intersystem crossing and electron excitation characteristics, resulting in significantly elevated reactive oxygen species (ROS) quantum yield. Our optimized lung-targeting nanoparticles enabled real-time NIR-II lung imaging and guided effective pulmonary coronavirus inactivation.

Organic single molecule based nano-platform for NIR-II imaging and chemo-photothermal synergistic treatment of tumor

A Renal-Clearable PEGylated Semiconducting Oligomer for the NIR-II Fluorescence Imaging of Tumor

Summary: Second near-infrared window fluorescence imaging (NIR-II FI) has gained significant attention in the field of bioimaging. However, most probes used for NIR-II FI undergo metabolization through the hepatobiliary pathway, which can lead to long-term toxicity. In this study, a renal-clearable semiconducting oligomer (PSO) was designed and synthesized for NIR-II FI of tumors. PSO consists of a semiconducting oligomer backbone as a fluorescence reporter and poly(ethylene glycol) side chains for water solubility. PSO emits an NIR-II fluorescence signal with a maximum emission at 1000 nm under excitation of 808 nm light. PSO exhibits good biocompatibility and can be partially cleared through renal clearance, making it a suitable probe for NIR-II FI of tumors as it effectively accumulates in the tumor.

Large Stokes shift benzothiazolium cyanine dyes with improved intramolecular charge transfer (ICT) for cell imaging applications

Summary: This article discusses the improvement of Stokes' shift in fluorescent dyes by introducing molecular asymmetry and the potential applications of these asymmetric dyes in live cell imaging. The optical properties of three asymmetric benzothiazolium dye designs are explored in detail. These new asymmetric ICT fluorophore systems exhibit large Stokes' shifts, high biocompatibility, wash-free staining, red to NIR emission, and easy excitation with commercially available laser wavelengths.
