Accurate Detection of Hcy in Human Serum and Two-Photon Visualization of Atherosclerosis Using a Highly Specific Fluorescent Probe

Summary: As a key metabolic intermediate for sulfur-containing amino acids, homocysteine (Hcy) is considered an independent risk factor for atherosclerotic cardiovascular disease. Therefore, real-time monitoring of Hcy levels is crucial for early diagnosis and treatment of atherosclerosis. A new two-photon fluorescent probe (RH-2) was developed using a hydrogen bond-assisted strategy, demonstrating high specificity for detecting Hcy in various biological samples. RH-2 successfully quantitatively determined Hcy in human serum and enabled two-photon fluorescence imaging of abnormal Hcy expression in atherosclerotic model mice, offering potential clinical applications for early diagnosis of atherosclerosis.

A Novel D-π-A Type Fluorescent Probe for Cu2+ Based on Styryl-Pyridinium Salts Conjugating Di-(2-picolyl)amine (DPA) Units

Summary: A novel D-pi-A type fluorescent probe L(NO3) for Cu(II) sensing was designed and characterized. The probe showed high selectivity for Cu2+ and its emission spectrum was strongly quenched upon binding. It exhibited strong anti-interference ability, excellent detection performance for Cu2+, and potential recycling properties. Moreover, inexpensive fluorescent test strips were designed for on-site and real-time Cu2+ detection with color recognition.

Summary: In this study, a novel copper complex, F-542-Cu2+, was developed for the detection of homocysteine (Hcy). F-542-Cu2+ showed significant reactivity towards biothiols and exhibited excellent environmental stability, superior sensitivity, and outstanding selectivity towards Hcy under physiological conditions. The study also demonstrated the potential application of F-542-Cu2+ in evaluating the activity of S-adenosylhomocysteine hydrolase (AHCY) both in vitro and in vivo.
