Quality assessment of Rheum species cultivated in Japan by focusing on M2 polarization of microglia

Summary: In traditional Japanese medicine, Rhei Rhizoma is used for its purgative, blood stasis-resolving, and antipsychotic properties, which may be related to its anti-inflammatory effects. This study investigated the effects of water extracts from different strains of cultivated Rheum species in Japan on microglia polarization. The results showed that the extracts of these strains significantly decreased M1 microglia and increased M2 microglia. Furthermore, certain constituents of Rhei Rhizoma were found to significantly increase the M2/M1 ratio.

Salvianolic acids from Salvia miltiorrhiza Bunge and their anti-inflammatory effects through the activation of a7nAchR signaling

Among them, C1 exhibited anti-inflammatory activities by activating a7nAchR signaling and subsequently inhibiting STAT3 and NF-KB pathways. Twist1 regulates macrophage plasticity to promote renal fibrosis through galectin-3

Summary: This study reveals a significant association between Twist1 expression in macrophages and the severity of renal fibrosis in IgA nephropathy patients and mice with unilateral ureteral obstruction. Twist1 ablation in macrophages alleviates renal tubular injury and fibrosis, accompanied by reduced macrophage infiltration and M2 polarization in the kidney. Twist1 downregulation inhibits macrophage chemotaxis and migration, at least partially through the CCL2/CCR2 axis. This study highlights the regulation of macrophage plasticity through Twist1/galectin-3 signaling and its potential as a new strategy for delaying kidney fibrosis in chronic kidney disease patients.

Sirt7 associates with ELK1 to participate in hyperglycemia memory and diabetic nephropathy via modulation of DAPK3 expression and endothelial inflammation

Summary: Diabetic nephropathy (DN) is a serious complication of advanced diabetes, and epigenetics-mediated hyperglycemic memory plays a crucial role in its pathogenesis. This study reveals that Sirt7 suppresses hyperglycemic memory in DN by modulating the expression of DAPK3 gene. Under high glucose conditions, decreased Sirt7 leads to increased DAPK3 expression and subsequent inflammatory response. In vivo experiments show that Sirt7 overexpression improves renal function and reduces inflammation in DN rats, while insulin treatment alone is ineffective.
