Adjuvant Therapy of Nivolumab Combined With Ipilimumab Versus Nivolumab Alone in Patients With Resected Stage IIIB-D or Stage IV Melanoma (CheckMate 915)

Jeffrey S. Weber et al.

Summary: The CheckMate 915 trial compared adjuvant nivolumab plus ipilimumab with nivolumab alone in patients with resected melanoma. The study found that there was no significant difference in recurrence-free survival between the two treatment groups.

Clinical characteristics and treatment outcomes of primary malignant melanoma of esophagus: a single center experience

Tae-Se Kim et al.

Summary: This study retrospectively analyzed the clinical characteristics and treatment outcomes of 17 patients with PMME. The results showed that radical resection surgery did not guarantee favorable treatment outcomes, and a novel treatment strategy is needed. Immunotherapy may have some efficacy for PMME patients, but further large-scale studies are warranted to confirm its effectiveness.

Y. Kurokawa et al.

Summary: This randomized, placebo-controlled, phase III trial showed that pimitespib significantly improved progression-free survival (PFS) and overall survival (OS) in patients with advanced gastrointestinal stromal tumors (GIST) refractory to standard tyrosine kinase inhibitors (TKIs), with an acceptable safety profile.

The Thousand Faces of Malignant Melanoma: A Systematic Review of the Primary Malignant Melanoma of the Esophagus

Gerardo Cazzato et al.

Summary: This article provides a systematic review of primary malignant melanoma of the esophagus (PMME) in terms of its history, epidemiology, treatment, and future research directions. PMME is an extremely rare form of esophageal cancer that has been subject to debate regarding its existence. Over time, more cases have been reported, increasing knowledge and paving the way for discussions on treatment. Currently, surgery remains the primary treatment approach, but new therapeutic strategies, such as nivolumab, are being explored. Future studies with large case series, including clinicopathological and molecular data, are needed to improve the outcome of PMME patients.

Mixed Neuroendocrine Non-Neuroendocrine Neoplasm Arising in the Ectopic Gastric Mucosa of Esophagus

Ryosuke Gushima et al.

Summary: Esophageal neuroendocrine neoplasms, especially mixed neuroendocrine non-neuroendocrine neoplasms (MiNENs), are extremely rare and have a poor prognosis. This case report describes a rare esophageal MiNEN, successfully treated with endoscopic submucosal dissection (ESD). The tumor originated from the ectopic gastric mucosa in the lower thoracic esophagus, and the patient remained cancer-free 24 months after the surgery.

Effectiveness of Etoposide and Cisplatin vs Irinotecan and Cisplatin Therapy for Patients With Advanced Neuroendocrine Carcinoma of the Digestive System The TOPIC-NEC Phase 3 Randomized Clinical Trial

Chigusa Morizane et al.

Summary: This study aimed to compare the efficacy of EP and IP in terms of overall survival (OS) in patients with NEC of the digestive system. The results showed no significant difference in OS between EP and IP, but EP had better outcomes in patients with poorly differentiated NEC of pancreatic origin. Adverse events were more common in the EP group.

Chigusa Morizane et al.

Summary: The results of this randomized clinical trial indicate that both EP and IP are still the standard first-line chemotherapy options, although grade 3 and 4 adverse events were more common in the EP arm.

Complete remission in metastatic primary malignant melanoma of the esophagus with nivolumab: a case report

Takeshi Okamoto et al.

Summary: Primary malignant melanoma of the esophagus is rare with poor prognosis. This case report demonstrates long-term complete remission of metastatic primary malignant melanoma of the esophagus after nivolumab monotherapy in a 79-year-old patient with a history of various cancers and comorbidities. Immune checkpoint inhibitors may have potential curative effects in selected populations, warranting further research to identify factors for achieving complete remission in primary malignant melanoma of the esophagus and other melanomas.

The management of metastatic GIST: current standard and investigational therapeutics

Ciara M. Kelly et al.

Summary: GIST is the most common mesenchymal tumor of the gastrointestinal tract, with KIT or PDGFR alpha mutations playing a key role in its development. Imatinib has revolutionized the management of GIST as first line treatment, but patients often experience disease progression after a few years of therapy. New treatment options are being explored for imatinib-refractory GIST.

Summary: In a study of advanced mucosal melanoma patients in Japan, it was found that there was no significant difference in efficacy between PD1 and PD1 + CTLA4, but the latter had a higher incidence of immune-related adverse events.

Esophageal Carcinosarcoma: Analysis of Clinical Features and Prognosis of 24 Cases and a Literature Review

Shusen Chen et al.

Summary: Esophageal carcinosarcoma (ECS) is a rare malignant tumor with controversial relationship between clinical features and prognosis. However, overall, ECS has a favorable prognosis, especially for patients with early stage who undergo radical esophagectomy. Further studies with more cases are needed to investigate this rare malignancy due to its low incidence.

Primary malignant melanoma of the esophagus successfully treated with nivolumab: a case report

Shingo Ito et al.

Summary: Primary malignant melanoma of the esophagus is a rare and aggressive disease with poor response to chemotherapy. This case study demonstrated successful treatment of recurrent primary malignant melanoma of the esophagus with the immune checkpoint inhibitor nivolumab, resulting in marked reduction of metastases and maintaining a partial response after 30 courses of treatment. No severe adverse events related to immunotherapy were reported, suggesting nivolumab may be a viable option for patients with metastatic primary malignant melanoma of the esophagus.
