Cyclodextrin-based tailored polyrotaxanes for highly efficient delivery of the genome-editing molecule

Off-target effects in CRISPR/Cas9 gene editing

Summary: Gene editing involves precise changes to specific nucleic acid sequences. The CRISPR/Cas9 system has revolutionized gene editing, making it efficient, convenient, and programmable. However, off-target effects remain a major concern, leading to unexpected alterations in the genome. Various methods have been developed to detect and mitigate these off-target effects, advancing the precision of CRISPR/Cas9 derivatives. This review summarizes these technological advancements and discusses the challenges in managing off-target effects for future gene therapy.

Versatile delivery platform for nucleic acids, negatively charged protein drugs, and genome-editing ribonucleoproteins using a multi-step transformable polyrotaxane

Summary: In this study, versatile and universal delivery carriers were developed using aminated polyrotaxane libraries for various biopharmaceuticals. The aminated polyrotaxane, which includes the carbamate bond between the axile molecule and endcap, was found to be suitable as a backbone polymer. The movable and flexible properties of the amino groups on polyrotaxane facilitated efficient complexation with different biopharmaceuticals, allowing for higher delivery efficacies without cytotoxicity than commercially available carriers.

Summary: RNA-guided systems play a central role in biological processes and provide adaptive immunity against foreign genetic elements. Recently, a new class of prokaryotic RNA-guided systems called OMEGA has been discovered, which includes the RNA-guided endonuclease TnpB. This study characterizes Fz, a eukaryotic OMEGA system, and demonstrates its potential for genome engineering applications in humans. The structure of Spizellomyces punctatus Fz is also resolved, revealing the conservation of core regions among Fz, TnpB, and Cas12.

Applying lessons learned from nanomedicines to understand rare hypersensitivity reactions to mRNA-based SARS-CoV-2 vaccines

Summary: After over a billion vaccinations with mRNA-LNP based SARS-CoV-2 vaccines, anaphylaxis and hypersensitivity reactions are considered rare adverse events. The underlying mechanisms of first-dose anaphylaxis and the predictability of future reactions are unknown. Given the importance of new mRNA constructs in addressing emerging viral variants, there is a need for safe repeated immunization approaches for high-risk individuals and reliable predictive tools for adverse reactions to mRNA vaccines. Lessons from nanomedicine research on infusion reactions can inform the mRNA vaccine field.

Current Cas9 RNP delivery systems, including physical approaches and synthetic carriers, are detailed in this review, along with their mechanisms and beneficial roles in intracellular delivery, highlighting examples of stimulus-responsive and targeted carriers in RNP delivery. Challenges of current Cas9 RNP delivery systems and perspectives in rational design of next generation materials for this promising field are discussed.
