Targeting BCAT1 Combined with a-Ketoglutarate Triggers Metabolic Synthetic Lethality in Glioblastoma

Bo Zhang et al.

Summary: This study reveals that BCAT1 plays a role in promoting the growth of IDHWT GBM. Loss of BCAT1 protein leads to the killing of IDHWT GBM cells by x-ketoglutarate (AKG), which can be reversed by reexpression of BCAT1 or supplementation with its downstream metabolic products, branched-chain ketoacids (BCKA). This finding uncovers a targetable metabolic vulnerability in the most common subset of GBM and provides a therapeutic strategy to improve glioblastoma treatment.

Regulation of branched-chain amino acid metabolism by hypoxia-inducible factor in glioblastoma

Bo Zhang et al.

Summary: This study reveals that hypoxia induces upregulation of the BCAA transporter LAT1 and the BCAA metabolic enzyme BCAT1 in human glioblastoma cells through the mediation of HIF-1 and HIF-2. HIF-1α plays a critical role in hypoxia-induced BCAT1 expression, and knockout of HIFs reduces glutamate labeling from BCAAs in GBM cells. Inhibition of BCAT1 inhibits GBM cell growth under hypoxia, highlighting a previously unrecognized HIF-dependent metabolic pathway.

Junjie Lv et al.

Summary: The study identified key genes related to survival in glioma, with PLAT, IGFBP2, BCAT1, and SERPINH1 showing potential as independent prognostic marker genes. These genes demonstrated good prognostic ability in distinct populations, highlighting the robustness and value of these prognostic markers.

Pathway-based classification of glioblastoma uncovers a mitochondrial subtype with therapeutic vulnerabilities

Luciano Garofano et al.

Summary: Garofano et al. classified glioblastomas into subtypes along metabolic and neurodevelopmental axes using single-cell RNA-sequencing data. Mitochondrial GBM showed the most favorable clinical outcome and vulnerability to inhibitors of oxidative phosphorylation, while glycolytic/plurimetabolic GBM relied on aerobic glycolysis and other metabolic pathways for energy production. This pathway-based classification can inform survival and precision targeting of cancer metabolism.
