TFEB-dependent lysosome biogenesis is required for senescence

Rachel Curnock et al.

Summary: The accumulation of senescent cells is a driver of aging, and one characteristic of senescent cells is an increase in lysosomal content. However, little is known about the causes and consequences of lysosomal biogenesis in senescence.

Lipofuscin Granule Accumulation Requires Autophagy Activation

Seon Beom Song et al.

Summary: This study examined the accumulation of lipofuscins in human fibroblasts under various stress conditions. The results showed that lipofuscins predominantly appear as granules that co-localize with autolysosomes in glucose-starved or replicative senescence cells, while they rarely manifest in the cytoplasm under oxidative stress induced by a doxorubicin pulse and chase. The findings suggest that autophagy activation triggers lipofuscin accumulation, and future treatments for lipofuscin-related diseases should consider autophagy activation.

S-Nitrosylation of p62 Inhibits Autophagic Flux to Promote α-Synuclein Secretion and Spread in Parkinson's Disease and Lewy Body Dementia

Chang-Ki Oh et al.

Summary: This article investigates the mechanism by which dysregulation of autophagic pathways leads to the accumulation of abnormal proteins and damaged organdies in neurodegenerative disorders. The authors found that pathologic protein S-nitrosylation of p62 is a critical factor for autophagic inhibition and cell-to-cell spread.

Long-lasting geroprotection from brief rapamycin treatment in early adulthood by persistently increased intestinal autophagy

Paula Juricic et al.

Summary: One challenge in repurposing the drug rapamycin for prolonging lifespan is to avoid side effects. The study shows that a brief early administration of rapamycin can have long-lasting beneficial effects in model organisms similar to lifelong treatment. This suggests that rapamycin has potential for geroprotection if side effects from continuous dosing can be avoided.

Glucose starvation induces autophagy via ULK1-mediated activation of PIKfyve in an AMPK-dependent manner

Cansu Karabiyik et al.

Summary: In response to glucose starvation, ULK1 activation by AMPK phosphorylates PIKfyve on S1548, leading to increased synthesis of the phospholipid PI(5)P and formation of PI(5)P-containing autophagosomes. This ULK1-mediated process enhances autophagy flux and reduces autophagy substrate levels.

Yahyah Aman et al.

Summary: This Review discusses the recent research on the mechanisms and roles of autophagy in health, aging, and disease, and how drugs modulating autophagy process could be used to suppress age-associated diseases. It examines emerging features of autophagy and proposes the potential links to aging and disease progression. The review also highlights the importance of understanding the interplay between autophagy and age-related pathologies for the development of clinical applications promoting long-term health.
