Self-assembly of photonic crystals by controlling the nucleation and growth of DNA-coated colloids

Alexander Hensley et al.

Summary: DNA-coated colloids can self-assemble into various crystal structures, and we have used microfluidics to quantify the kinetics of crystallization. Our study shows that classical theories can accurately predict nucleation and growth rates, and we have successfully designed large single crystals with specific structural coloration.

Manipulating polydispersity of lens beta-crystallins using divalent cations demonstrates evidence of calcium regulation

Michael R. Bergman et al.

Summary: This study investigates the polydispersity of beta-crystallins and reveals that it is determined by isoform composition and the presence of divalent cationic salts. High concentrations of divalent cations can dissociate beta-crystallin oligomers, reduce polydispersity, and alter the protein surface charge. This dynamic process may play a crucial role in the molecular packing and optical function of the lens.

Insights into the biochemical and biophysical mechanisms mediating the longevity of the transparent optics of the lens

Roy A. Quinlan et al.

Summary: In this review, the authors highlight the potential protective effects of post-translational modifications (PTMs) on lens proteins during normal development and propose a forward-looking hypothesis that aims to prevent age-related cataract (ARC) by protecting the biochemical and biophysical properties of lens proteins.

Macromolecular Crowding and Intrinsically Disordered Proteins: A Polymer Physics Perspective

Jasmine Cubuk et al.

Summary: This article discusses the effects of cellular environment on intrinsically disordered proteins. Unlike structured proteins, intrinsically disordered proteins do not have a stable three-dimensional structure and remain flexible and dynamic in solution. The physics of polymers and colloids provides a framework to interpret how crowding affects the conformation, dynamics, and interactions of disordered proteins.

Imbalances in the eye lens proteome are linked to cataract formation

Philipp W. N. Schmid et al.

Summary: Analysis of lenses from different mouse strains with early-onset cataracts due to mutations in alpha-, beta-, or gamma-crystallin proteins reveals that the mutant crystallins are unstable in vitro, reduced in levels within the lens, and do not accumulate in the water-insoluble fraction. Precipitation of other crystallin proteins, including alpha-crystallins, suggests that imbalance in the lenticular proteome and altered crystallin interactions are the basis for cataract formation, rather than the aggregation propensity of the mutant crystallins.

Impact of In-Cell and In-Vitro Crowding on the Conformations and Dynamics of an Intrinsically Disordered Protein

Iwo Konig et al.

Summary: The concentrations of macromolecules within cells can affect the conformations and dynamics of proteins. By reducing cell volume through hyperosmotic stress, it was found that intrinsically disordered proteins exhibit compaction and slower dynamics in more crowded cells. This concentration and length-scale dependence of crowding can be explained by polymer theory and depletion interactions.

Analysis of amyloid-like secondary structure in the Cryab-R120G knock-in mouse model of hereditary cataracts by two-dimensional infrared spectroscopy

Ariel M. Alperstein et al.

Summary: Alpha B-crystallin is a small heat shock protein that can form aggregates in cataracts and Alzheimer's disease. The R120G mutation in this protein is associated with hereditary cataracts and myopathy, and studying the secondary structures of these aggregates is important for understanding disease mechanisms and developing treatments. The 2DIR spectroscopy study confirmed the presence of amyloid-like secondary structures in Cryab-R120G knock-in mice with cataracts, supporting the use of this model for studying age-related cataract disease.
