Photoredox/HAT-Catalyzed Dearomative Nucleophilic Addition of the CO2 Radical Anion to (Hetero)Aromatics

Summary: The radical anion of CO2 is a highly nucleophilic radical species with many potential applications in organic chemistry. In this study, a robust method for introducing CO2 center dot- into stable heteroaromatics was developed. The resulting reactions produced synthetically useful alpha-oxy, alpha-thio, and alpha-amino acid derivatives, as well as carboxylated tetrahydronaphthalene derivatives. These dearomative carboxylation reactions are unprecedented and provide a synthetic option for introducing a C1 source into stable (hetero)aromatics.

Translating Planar Heterocycles into Three-Dimensional Analogs by Photoinduced Hydrocarboxylation

Summary: The rapid preparation of complex three-dimensional heterocyclic scaffolds is a crucial challenge in medicinal chemistry. Despite the dominance of flat molecules in new drug targets due to the availability of coupling reactions, a new strategy has been reported to enable the transformation of readily accessible planar molecules into more three-dimensionally complex analogs through dearomative hydrocarboxylation of indoles and related heterocycles. This highly chemoselective, broad in scope, operationally simple, and readily amenable to high-throughput experimentation (HTE) reaction will allow the translation of existing libraries of heteroaromatic compounds into diverse 3D analogs and exploration of new classes of medicinally relevant molecules.

Mechanochemical Approach for Air-Tolerant and Extremely Fast Lithium-Based Birch Reductions in Minutes

We report an ammonia-free lithium-based Birch reduction using ball-milling technique, which is characterized by its simplicity, short reaction time, broad substrate scope, and no requirement for dry bulk solvents. The method shows potential in reducing bioactive target molecules and gram-scale synthesis.

A General Light-Driven Organocatalytic Platform for the Activation of Inert Substrates

Summary: Activating inert substrates is challenging due to their strong covalent bonds and low reduction potentials. Recent advances in photoredox catalysis have provided solutions to activate specific inert bonds. However, developing a general catalytic platform that can consistently target a broad range of inert substrates would be synthetically useful. This study reports the use of an indole thiolate organocatalyst that, when excited with 405 nm light, becomes strongly reducing and able to activate strong C-F, C-Cl, and C-O bonds in both aromatic and aliphatic substrates. Mechanistic studies revealed an excited-state thiolate anion responsible for the highly reducing reactivity.

Radical Hydrocarboxylation of Unactivated Alkenes via Photocatalytic Formate Activation

Summary: In this study, we propose a strategy to promote the hydrocarboxylation of unactivated alkenes using photochemical activation of formate salts. We demonstrate an alternative initiation mechanism that overcomes the limitations of previous approaches and enables effective hydrocarboxylation of challenging substrate class. By accessing the necessary thiyl radical initiator without an exogenous chromophore, we can eliminate the major byproducts that hindered similar reactivity for unactivated alkene substrates. This redox-neutral method is simple to execute and applicable to a wide range of alkene substrates, including feedstock alkenes like ethylene, which can be hydrocarboxylated under ambient conditions. Further radical cyclization experiments explore the potential redirection of the described reactivity through more complex radical processes.

Photocatalytic phosphine-mediated water activation for radical hydrogenation

Summary: Chemical activation of water is a hot topic in energy research and enables the conversion of this abundant resource into valuable compounds. In this study, we demonstrate the activation of water using a photocatalytic phosphine-mediated radical process, which involves sequential heterolytic and homolytic cleavage of O-H bonds. The resulting PR3-OH radical intermediate can react with various closed-shell p systems, leading to transfer hydrogenation of the p system and incorporation of hydrogen atoms from water into the product. Experimental and theoretical studies support the importance of the hydrogen atom transfer step in this radical hydrogenation process.

Mild and metal-free Birch-type hydrogenation of (hetero)arenes with boron carbonitride in water

Summary: This study presents a system based on boron carbonitride semiconductor that can reduce inert planar arenes and heteroarenes to water under blue light irradiation. The system offers the advantages of low-cost, high efficiency, easy separation, environmentally friendly profile, and broad substituent tolerance. It can be applied to late-stage functionalization and/or deuteration of drugs, hormones, and axially chiral compounds while conserving their chirality.
