GTS-21 inhibited inflammation by reducing the expression of pro-inflammatory cytokines and activating anti-inflammatory signaling pathways. It also showed neuroprotective properties by reducing microglial activation and protecting dopaminergic neurons.

In the cell model, miR-142-5p is significantly upregulated by muscone treatment, and downregulation of miR-142-5p eliminates the neuroprotective effects of muscone against cerebral hypoxia injury.

Proteomics on the role of muscone in the consciousness-restoring resuscitation effect of musk on ischemic stroke

Summary: This study aimed to identify the target of muscone in the treatment of ischemic stroke using network pharmacology, an animal model of ischemic stroke, and differential proteomics. The results showed that muscone can reduce neuronal necrosis, protect the blood-brain barrier, and improve neurological damage caused by cerebral ischemia mainly through the regulation of neuronal synaptic connections. Muscone is an important active component responsible for the consciousness-restoring resuscitation effect of musk on ischemic stroke.

Summary: Impairments in mitochondrial physiology, specifically the muscarinic acetylcholine type 1 receptor (M1R) pathway, play a role in the progression of neurodegenerative conditions such as peripheral neuropathy in diabetes. Inhibition of M1R using specific antagonists can prevent mitochondrial dysfunction and nerve degeneration. Activation of AMPK through pirenzepine and muscarinic toxin 7 (MT7) can improve sensory abnormalities and nerve fiber loss. The negative regulation of the M1R-dependent pathway could be a potential therapeutic target for enhancing mitochondrial function and repairing nerve damage.
