Cu-GA-coordination polymer nanozymes with triple enzymatic activity for wound disinfection and accelerated wound healing

Haotian Tian et al.

Summary: In recent years, bacterial infection and oxidative stress have posed significant challenges to wound healing. The rise of drug-resistant superbugs has particularly hindered the treatment of infected wounds. The development of new nanomaterials, such as the coordination polymer copper-gallic acid (Cu-GA) nanorods with multi-enzyme activity, has emerged as a promising approach for the treatment of drug-resistant bacterial infections. Cu-GA exhibits enhanced multi-enzyme activity under different pH conditions, effectively killing bacteria in acidic conditions while promoting wound healing in neutral conditions. Furthermore, in vivo studies demonstrate that Cu-GA promotes wound infection healing with good biosafety.

Biological Glue from Only Lipoic Acid for Scarless Wound Healing by Anti-inflammation and TGF-? Regulation

Caikun Liu et al.

Summary: LA-glue, a new type of bioglue, can effectively seal wounds noninvasively and has great potential in scarless wound healing. Compared with current bioglues, LA-glue can regulate inflammatory cytokines and TGF-fi1, two key factors in scar formation. Experimental results showed that LA-glue treated wounds not only had the same appearance as normal skin, but also had restored tensile strength and hair follicle density. This simple and safe LA-glue addresses the three key factors of scar formation, making it promising for clinical applications.

Emergence of Plant-Based Decellularized Scaffolds for Tissue Regeneration: A Review

Jagadeesh Kumar Reddy Mutra et al.

Summary: A scaffold is necessary in tissue engineering to support cell adhesion and proliferation by mimicking the targeted injury site and allowing nutrient and growth factor exchange. Current methods involve making synthetic or biocomposite scaffolds using polymers or extracting natural polymers from plants or animals. This review focuses on the direct use of natural materials for scaffold preparation and explores plant materials that can be used. Decellularization is found to be a commonly used method, and various characterization techniques such as swelling nature, biodegradability, porosity, surface morphology, and mechanical strength are discussed. Analytical techniques like SEM, FT-IR, and UV spectroscopy are also explored for chemical analysis and cell adherence determination.

Integrating zinc/silicon dual ions with 3D-printed GelMA hydrogel promotes in situ hair follicle regeneration

Fanliang Zhang et al.

Summary: This study investigated the synergistic effects of zinc and silicon ions on dermal cells and determined the optimal concentration for medical applications. A scaffold was created by integrating zinc/silicon dual ions into GelMA, and it was found to have suitable mechanical properties for biological treatment. The scaffold was then used to promote in situ hair follicle regeneration in a mouse excisional model, showing significant activation of hair follicle stem cells and enhanced neovascularization.

ROS-responsive microneedles loaded with integrin av beta 6-blocking antibodies for the treatment of pulmonary fibrosis

Hao Ding et al.

Summary: We have developed a micro-invasive percutaneous and hydrogen peroxide-responsive microneedle for smart delivery of integrin alpha v beta 6-blocking antibody. This microneedle has advantages of rapid response, excellent biocompatibility, protection of bioactivity, high tissue permeation, and specific targeting to lesions. It can partially release the antibody when exposed to hydrogen peroxide generated during pulmonary fibrosis, reducing activation of the pro-fibrotic factor TGF-beta 1 and showing excellent therapeutic efficacy.

Injectable Reactive Oxygen Species-Responsive Hydrogel Dressing with Sustained Nitric Oxide Release for Bacterial Ablation and Wound Healing

Jie Yu et al.

Summary: This study establishes a system consisting of a degradable hydrogel dressing that reacts to reactive oxygen species, an encapsulated biosafe nitric oxide donor, and a low concentration of hydrogen peroxide as a disinfectant. This system promotes sustainable wound healing and disinfection by generating nitric oxide and mediating cell migration and collagen synthesis. It also demonstrates synergistic antibacterial properties against antibiotic-resistant bacteria.

Copper Clusters: An Effective Antibacterial for Eradicating Multidrug-Resistant Bacterial Infection In Vitro and In Vivo

Xiangchun Zhang et al.

Summary: CuCs is a novel antibacterial molecule with broad-spectrum and excellent antibacterial activity, able to disrupt bacterial wall structure, regulate GSH/GSSG ratio, induce reactive oxygen species production, and ultimately lead to bacterial death. In vivo studies in mice demonstrate that CuCs can significantly treat skin wound infections and sepsis caused by MRSA, with similar therapeutic efficacy as mupirocin ointment and vancomycin. Compared to silver and platinum clusters, CuCs show extremely low cytotoxicity to normal mammalian cells.

Using bioprinting and spheroid culture to create a skin model with sweat glands and hair follicles

Yijie Zhang et al.

Summary: An in vitro model of skin constructs with sweat glands (SGs) and hair follicles (HFs) was established to explore the interaction between these two appendages in regeneration. The results showed that HF spheroids promoted differentiation of both SGs and HFs in SG scaffolds, while SG scaffolds mainly promoted SG differentiation and had little effect on HF potency in HF spheroids. The study provides insights into the molecular mechanisms of SG and HF regeneration.
