Stellettin B Induces Cell Death in Bladder Cancer Via Activating the Autophagy/DAPK2/Apoptosis Signaling Cascade

Chun-Han Chang et al.

Summary: Bladder cancer (BC) is a common and urgent medical issue, yet the current treatment approaches have limited effectiveness. This study investigated the anti-BC activity of SP-2, a triterpene isolated from a marine sponge. SP-2 suppressed BC cell viability and induced apoptosis through the inhibition of the FGFR3-TACC3/Akt/mTOR pathway, and it also triggered autophagy and upregulated DAPK2 expression to enhance cytotoxicity and apoptosis.

KRAS Mutations in Solid Tumors: Characteristics, Current Therapeutic Strategy, and Potential Treatment Exploration

Yunkai Yang et al.

Summary: The KRAS gene is commonly mutated in solid tumors, but targeting it has been a challenge. However, recent breakthroughs with KRAS G12C inhibitors show promise in targeted therapy, although resistance may occur. Therefore, non-targeted conventional therapies are also considered promising.

Apoptotic and antimetastatic effect of cucurbitacins in cancer: recent trends and advancement

Ajay Kumar et al.

Summary: Cucurbitacins, a class of secondary metabolites produced by the Cucurbitaceae family, have significant anticancer activity by inhibiting cell proliferation, invasion, and migration, inducing apoptosis, and encouraging cell cycle arrest. They also suppress the JAK-STAT3, Wnt, PI3K/Akt, and MAPK signaling pathways, which are essential for cancer cell survival and apoptosis. This study provides a comprehensive summary of potential molecular targets for cucurbitacins in suppressing various malignant processes.

Synergistic effect of cucurbitacin E and myricetin on Anti-Non-Small cell lung cancer: Molecular mechanism and therapeutic potential

Jinfang Zhang et al.

Summary: This study explored the therapeutic effect of the combination of Cucurbitacin E (CuE) and Myricetin (Myr) from the herb Citrullus colocynthis on non-small cell lung cancer (NSCLC). The results showed that this combination inhibited cancer cell proliferation and induced apoptosis, as well as suppressed autophagy and the activation of the PI3K/AKT/mTOR signaling pathway.

Summary: The American Cancer Society predicts that there will be 1,958,310 new cancer cases and 609,820 cancer deaths in the United States in 2023. While incidence trends for most cancers are favorable, prostate cancer saw a significant increase of 3% annually from 2014 to 2019. However, the overall cancer death rate continues to decline due to advances in treatment, with a 33% reduction since 1991.

Targeted Therapies for Lung Cancer Patients With Oncogenic Driver Molecular Alterations

Aaron C. Tan et al.

Summary: The therapeutic landscape of non-small-cell lung cancer is undergoing significant changes with the shift from histology-based classification to molecularly defined subsets. Targeted therapies for specific driver mutations such as EGFR, ALK, and ROS1 have shown promising results, and there is a growing list of approved therapies for other mutations. The importance of diagnostic molecular testing, optimal sequencing of therapies, application of targeted therapies in early-stage disease, and the impact of genomic mutations on targeted therapy efficacy are important areas of research.

The Tubulin Code and Tubulin-Modifying Enzymes in Autophagy and Cancer

Daniela Trisciuoglio et al.

Summary: This review focuses on the significant role of tubulin modifications in determining different cancer characteristics, including metastatic cell migration, therapy resistance, and autophagy. It highlights the potential of inducing microtubule modifications as a means to kill cancer cells and the influence of modulating tubulin-modifying enzymes on cancer cell survival and aggressiveness. It suggests that targeting tubulin post-translational modifications can be a promising avenue in cancer therapy.

Cucurbitacins as potential anticancer agents: new insights on molecular mechanisms

Carla Varela et al.

Summary: Cucurbitacins, found primarily in the Cucurbitaceae family but also in other plant families, fungi, and a marine mollusc, have significant biological activities such as anti-inflammatory, antioxidant, antimalarial, antimicrobial, hepatoprotective, and anticancer potential. Chemical synthesis of cucurbitacins has been developed due to limitations in their natural obtaining and isolation. Comprehensive reviews on this topic are recommended for future clinical research.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Daniel J. Klionsky et al.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
