Critical Evaluation of Different Lysosomal Labeling Methods Used to Analyze RNA Nanocarrier Trafficking in Cells

Shoaib Iqbal et al.

Summary: The field of using nucleic acids to regulate gene expression has immense clinical potential. Nanomaterials are often used to deliver nucleic acids into cells, and fluorescent endo/lysosomal markers are commonly employed tools to evaluate the endosomal escape of nucleic acid nanocarriers. These markers can alter cellular function and cell-nanocarrier interactions, affecting nanoparticle trafficking studies.

Polycation-Mediated Transfection: Mechanisms of Internalization and Intracellular Trafficking

Summary: Polyplex-mediated gene transfection is complex, with untargeted polyplexes not following traditional mechanisms and targeting syndecans. Endocytosis requires free polycations to disrupt cell membranes and promote polyplex escape. Polyplexes release plasmids into the nucleus from the endosome, but only a small proportion of plasmids ultimately express.
