Design, synthesis, in silico ADME, DFT, molecular dynamics simulation, anti-tyrosinase, and antioxidant activity of some of the 3-hydroxypyridin-4-one hybrids in combination with acylhydrazone derivatives

Summary: In this study, hybrids of 3-hydroxypyridine-4-one and acylhydrazones were synthesized and evaluated for their anti-tyrosinase and antioxidant activities. The results showed that compound 5h exhibited potent inhibitory effects on tyrosinase and strong antioxidant activity. Molecular docking and dynamics simulation confirmed the binding site of compound 5h with tyrosinase. Furthermore, reactivity descriptors and molecular orbitals of 5g and 5h were investigated using DFT calculations and compared with experimental data.

Effect of ascorbic acid on tyrosinase and its anti-browning activity in fresh-cut Fuji apple

Summary: This research investigated the impact of ascorbic acid on tyrosinase and its anti-browning effect on fresh-cut Fuji apple. Ascorbic acid demonstrated a dual effect on tyrosinase with an IC50 of 13.40 +/- 0.05 mu M, and it prevented browning by interacting with the enzyme and increasing APX activity while inhibiting PPO and POD activities. These findings suggest a feasible application of ascorbic acid for fruit preservation.

The basicity of an active-site water molecule discriminates between tyrosinase and catechol oxidase activity

Summary: The study showed that the presence of metallochaperone caddie protein (Cad) enables tyrosinase (Ty) to catalyze the catecholase reaction more effectively than the phenolase reaction. Mutations at the N191 residue can decrease the inherent ratio of phenolase versus catecholase activity. The increased basicity of the water molecule may be necessary for inhibiting the proton transfer in the catalytic cycle.
