A robust ratiometric photoacoustic and fluorescence dual-modal molecular probe for capturing endogenous HNO generation in tumor mice model

Summary: In this study, the first near-infrared PA/FL dual-modal probe DOP-HNO was designed and synthesized. It specifically and sensitively responded to HNO in vivo, successfully visualized HNO fluctuations in cells and living mice, and observed the interaction between NO and H2S in the tumors of living mice through NIR dual-ratiometric PA/FL imaging. For the first time, it provided visualization evidence of endogenous HNO generation in a cancer model and investigated its possible generation mechanisms. Therefore, the dual-modal probe DOP-HNO is an effective tool to study the complex relationship between HNO and cancer as well as the pathology of other HNO-related diseases.

In Situ Synthesis of AIEgen-based Porous Organic Polymer Films by Interfacial Amino-yne Click Polymerization for Efficient Light-Harvesting

Summary: We have developed a catalyst-free, atom-economical interfacial amino-yne click polymerization method for the synthesis of aggregation-induced emission luminogen (AIEgen)-based free-standing porous organic polymer (POP) films at room temperature. These films exhibited excellent crystalline properties and good porosity as confirmed by X-ray diffraction and N-2 uptake experiments, respectively. The thickness of the POP films could be easily controlled by adjusting the monomer concentration. Furthermore, these AIEgen-based POP films demonstrated high luminescence with a photoluminescent quantum yield of up to 37.8% and displayed good chemical and thermal stability. Moreover, the introduction of an organic dye into the POP film allowed for the formation of an artificial light-harvesting system with a large red-shift, highly efficient energy-transfer ability, and high antenna effect.

Unexpected dual-functional cyanine fluorophores: NIR multifunctional fluorescent probes for simultaneous monitoring of mitochondrial cysteine /viscosity during kidney ferroptosis in vivo

Summary: This study developed a series of novel cyanine fluorophores for simultaneous visualization of cysteine level and mitochondrial viscosity during kidney ferroptosis. The results showed that ferroptosis process was accompanied by a decrease in cysteine channel and an increase in mitochondrial viscosity channel. Furthermore, the study also revealed dynamic changes of cysteine level and mitochondrial viscosity in a mouse kidney ferroptosis model in vivo.

A General Approach to Design Dual Ratiometric Fluorescent and Photoacoustic Probes for Quantitatively Visualizing Tumor Hypoxia Levels In Vivo

Summary: This study describes an energy balance strategy by sulfur substitution to optimize hemicyanine dyes for dual ratiometric fluorescence and photoacoustic imaging. The newly developed nitroreductase probe AS-Cy-NO2 allows quantitative visualization of tumor hypoxia in vivo with high sensitivity and depth. This promising NIRF/PA scaffold may have broader applications in dual ratiometric imaging of other disease-relevant biomarkers.

Summary: This study reported a novel dual ratiometric probe DOP-CO for reliable imaging and quantitative detection of carbon monoxide (CO) in live animals. The probe showed successful application in imaging the release behavior of exogenous and endogenous CO and provided valuable insights into the relationship between CO fluctuations and liver injury. It also offers a promising tool for investigating more CO-mediated biological processes.
