Carboxymethyl chitosan/sodium alginate hydrogels with polydopamine coatings as promising dressings for eliminating biofilm and multidrug-resistant bacteria induced wound healing

Summary: A mussel-inspired multifunctional hydrogel (CAC/PDA/Cu(H2O2)) was prepared, which exhibited antibacterial, anti-inflammatory, and angiogenesis-promoting properties, accelerating wound healing. The hydrogel showed excellent therapeutic effects in Methicillin-resistant Staphylococcus aureus (MRSA) induced-rat infection models, efficiently eliminating MRSA, significantly reducing inflammatory expression, promoting angiogenesis, and ultimately shortening the wound healing time.

Controlled release of protein from gelatin/chitosan hydrogel containing platelet-rich fibrin encapsulated in chitosan nanoparticles for accelerated wound healing in an animal model

Summary: In this study, platelet-rich fibrin-chitosan (CH-PRF) nanoparticles were loaded into gelatin-chitosan hydrogel (Gel-CH/CH-PRF) to create a hydrogel dressing for wound healing. The Gel-CH/CH-PRF hydrogel exhibited excellent swelling behavior, good porosity, high absorption of wound exudates, and proper vapor permeability rate. It also demonstrated sustained release of proteins and strong antimicrobial activity. In vivo experiments showed that the Gel-CH/CH-PRF hydrogel promoted wound closure, increased epidermis thickness, and enhanced the formation of new granulation tissue. These findings highlight the potential of Gel-CH/CH-PRF hydrogel as an effective wound dressing.

Antibacterial, photothermal and stable Ag-titanium-oxo-clusters hydrogel designed for wound healing

Summary: In this study, a stable and highly effective antibacterial and anti-inflammatory Ag-Titanium-oxo-cluster (Ag-TOC, Ag9Ti4) was developed through one-step solvothermal method by introducing silver and salicylic acid. The Ag9Ti4 cluster was incorporated into a dopamine-containing hydrogel system, which exhibited excellent antibacterial and photothermal properties in vitro. Furthermore, the Ag9Ti4 hydrogel system demonstrated superior anti-inflammatory and wound healing ability under NIR condition in vivo. The construction of the antibacterial, photothermal, and stable Ag-TOC hydrogel system provides a new strategy for expanding the biomedical applications of TOCs.

A hydrogel system containing molybdenum-based nanomaterials for wound healing

Summary: Delayed or non-healing wounds have a significant impact on patients' quality of life and pose a financial burden on public health systems worldwide. A hydrogel containing low-valence-state molybdenum nanomaterials has been developed to scavenge reactive oxygen species (ROS) and promote wound healing. In vitro and in vivo studies have shown that this novel Mo hydrogel accelerates wound healing, stimulates angiogenesis, and enhances growth factor expression. Due to the essentiality of molybdenum in all organisms, this Mo hydrogel holds great potential for clinical translation in wound healing applications.

An antioxidant and antibacterial polydopamine-modified thermo-sensitive hydrogel dressing for Staphylococcus aureus-infected wound healing

Summary: In this study, a thermo-sensitive and ROS-scavenging hydrogel based on polydopamine modified poly(epsilon-caprolactone-co-glycolide)-b-poly(ethylene glycol)-b-poly(epsilon-caprolactone-co-glycolide) (PDA/P2) triblock copolymer was developed. The hydrogel showed good ROS-scavenging ability and antibacterial activity against E. coli and S. aureus. In vivo experiments demonstrated that the hydrogel dressing promoted wound healing by inhibiting bacterial growth, alleviating inflammation, and inducing angiogenesis and collagen deposition.

Multifunctional MOF-Based Microneedle Patch With Synergistic Chemo-Photodynamic Antibacterial Effect and Sustained Release of Growth Factor for Chronic Wound Healing

Summary: Researchers have developed a multifunctional microneedle (MN) patch that achieves rapid wound healing through efficient chemo-photodynamic antibacterial effects and sustained release of growth factors. The MN patch contains low-dose antibiotics and metal-organic frameworks (MOFs) encapsulating bioactive small molecules. When the MN patch penetrates the skin, the drugs and bioactive substances are rapidly released to the wound. Under light irradiation, MOF-based nanoparticles convert O-2 into O-1(2), synergistically removing pathogenic bacteria from the wound. The nanoparticles also continuously release growth factors, promoting the formation of epithelial tissue and neovascularization to accelerate chronic wound healing.

An all-natural hydrogel has been developed, which can convert M1 macrophages into M2 macrophages without additional ingredients or external intervention, providing a simple and safe approach for accelerating wound healing.

Hypoxia-pretreated ADSC-derived exosome-embedded hydrogels promote angiogenesis and accelerate diabetic wound healing

Summary: Adipose-derived stem cell (ADSC)-originating exosomes (ADSC-Exos) have potential in diabetic wound repair, but their disadvantages limit their clinical application. This study developed hypoxia-pretreated ADSC-Exo (ADSC-HExo)-embedded GelMA hydrogels (GelMA-HExo) and demonstrated their therapeutic effects in promoting diabetic wound healing. These findings suggest a new therapeutic strategy for diabetic ulcers.

The use of nanomaterials has shown promise in promoting wound healing due to their unique physicochemical characteristics at the nanoscale. This review provides an overview of wound categories, the wound healing process, and critical influencing factors, and discusses the applications of nanomaterials with intrinsic therapeutic effects and nanomaterials-based drug delivery systems in promoting wound healing. Limitations and future perspectives of nanomaterials in wound healing are also discussed.

Intelligent microneedle patch with prolonged local release of hydrogen and magnesium ions for diabetic wound healing

Summary: Diabetes mellitus leads to delayed wound healing, inflammation, decreased angiogenesis, and increased reactive oxygen species. A microneedle patch made of PLGA and loaded with MgH2 is developed to address diabetic wounds. The patch delivers MgH2 transdermally, which reacts with body fluids to release hydrogen and magnesium ions. The released hydrogen reduces reactive oxygen species, transforming the pathological microenvironment, while the magnesium ions promote the polarization of pro-healing M2 macrophages. This intelligent microneedle patch provides a novel approach for accelerating wound healing through local and sustainable preservation and release of H2 and Mg2+.

Further in vivo experiments are needed to assess the clinical efficacy of the hydrogel films.

Summary: We developed biocompatible and nonirritating triple-helical recombinant collagen (THRC) dressings for accelerated healing of microneedle-injured and photodamaged acute skin wounds. The THRC dressings exhibited superior biocompatibility and bioactivity, significantly promoting the proliferation and adhesion of fibroblasts. In vivo studies demonstrated that the THRC dressings effectively healed damaged dermis and accelerated re-epithelialization and collagen deposition.
