Data-Independent Acquisition Phosphoproteomics of Urinary Extracellular Vesicles Enables Renal Cell Carcinoma Grade Differentiation

Marco Hadisurya et al.

Summary: The translation of cancer signaling research into clinical application has been slow and ineffective. However, extracellular vesicles (EVs) show promise as a source for developing disease phosphoprotein markers. This study focuses on using mass spectrometry to profile urinary EV phosphoproteomics for renal cell cancer (RCC) differentiation. By optimizing the process of EV isolation, phosphopeptide enrichment, and DIA method, this study provides a powerful tool for future clinical applications.

Microfluidic Platform for Profiling of Extracellular Vesicles from Single Breast Cancer Cells

Jonas M. Nikoloff et al.

Summary: By analyzing the protein profiles of single extracellular vesicles (EVs), we found that EVs released by a single cell can represent its cell of origin. The microfluidic platform we used allowed for immobilization of EVs from individual cells, reducing the heterogeneity seen in cell populations. Through immunostaining and fluorescence microscopy, we determined the biochemical characteristics of membrane and cytosolic proteins in EVs. Our study demonstrates the potential of EVs as diagnostic markers and provides a method for studying EV biogenesis.

One-Pot Analytical Pipeline for Efficient and Sensitive Proteomic Analysis of Extracellular Vesicles

Yi-Kai Liu et al.

Summary: Extracellular vesicle proteomics is a useful tool for discovering biomarkers for disease diagnosis and treatment. However, the current workflow for analyzing EV proteomes using mass spectrometry is not suitable for clinical settings. In this study, we developed a one-pot analytical pipeline that combines a robust EV isolation method and in situ protein sample preparation to detect urinary EV proteomes. This streamlined pipeline enabled the analysis of the entire EV proteome in one day and yielded better results compared to existing methods.

Exosomes as a new frontier of cancer liquid biopsy

Dan Yu et al.

Summary: Liquid biopsy, a revolutionary strategy for cancer diagnosis and prognosis prediction, uses minimally invasive detection through biofluids such as blood, saliva, and urine. Exosomes, a subset of extracellular vesicles, play a crucial role in intercellular communication and are considered as potential biomarkers in liquid biopsy. However, the lack of high-quality separation and component analysis methods has limited the further clinical application of exosomes.

Role of Serine Proteases at the Tumor-Stroma Interface

Ravichandra Tagirasa et al.

Summary: The interaction between tumor and stroma during tumor development plays a crucial role in tumor progression, therapy, and immune response. Proteolytic enzymes, particularly serine proteases, expressed by tumor and stromal cells, are important in modulating tumor-stroma interaction. This review highlights the role of stromal-derived serine proteases in tumor progression and their potential therapeutic applications.

Insight into Extracellular Vesicle-Cell Communication: From Cell Recognition to Intracellular Fate

Lana Ginini et al.

Summary: Extracellular vesicles (EVs) are crucial heterogenous lipid structures secreted by cells for cell-to-cell communication. EVs primarily enter cells through endocytosis, a process that may depend on proteins and lipids on the surfaces of both vesicles and target cells.
