Discovery of novel cannabidiol derivatives with augmented antibacterial agents against methicillin-resistant Staphylococcus aureus

Benzenesulfonyl thiazoloimines as unique multitargeting antibacterial agents towards Enterococcus faecalis

Summary: Newly developed benzene-sulfonyl thiazoloimines (BSTIs) with unique structure have shown promising antibacterial potential against drug-resistant pathogens, with 2-pyridyl BSTI 14d being particularly effective against E. faecalis. Compound 14d exhibited rapid bactericidal properties, inhibited biofilm formation, and displayed low toxicity towards human cells. Mechanistic investigations revealed that compound 14d disrupted cell membranes, caused oxidative damage, and hindered DNA replication. These findings suggest that BSTIs could provide a new possibility for overcoming drug resistance.

New Efforts toward Aminothiazolylquinolones with Multitargeting Antibacterial Potential

Summary: New antibacterial 3-(aminothiazolyl)quinolones were synthesized to combat multidrug resistance in animal husbandry. Bioactive assays showed that N,N-dicyclohexylaminocarbonyl ATQ 10e and methyl ATQ 17a exhibited better antibacterial behavior against Staphylococcus aureus and Pseudomonas aeruginosa than norfloxacin. Mechanism exploration indicated that ATQ 17a could inhibit biofilm formation, disrupt bacterial membrane integrity, bind to intracellular DNA and DNA gyrase, induce reactive oxygen species production, and reduce bacterial metabolism.

Discovery of Amphiphilic Xanthohumol Derivatives as Membrane-Targeting Antimicrobials against Methicillin-Resistant Staphylococcus aureus

Summary: Infections caused by multidrug-resistant (MDR) bacteria are increasing worldwide, and novel antimicrobials are urgently needed. In this study, a class of amphiphilic xanthohumol derivatives were synthesized and molecule IV15 showed remarkable antibacterial effects, rapid bactericidal properties, low toxicity, good plasma stability, and reduced bacterial resistance. Mechanistic studies revealed that IV15 disrupted bacterial cell membranes, leading to bacterial death by increasing reactive oxygen species and causing leakage of proteins and DNA. Importantly, IV15 exhibited superior in vivo anti-MRSA efficacy compared to vancomycin.

Summary: Cannabidiol (CBD) from Cannabis sativa is used in cosmetics due to its antibacterial and antioxidant properties, and a non-intoxicating CBD alternative called 8,9-dihydrocannabidiol (H2CBD) has been synthesized and studied. H2CBD shows strong antibacterial and antioxidant activity, as well as lower toxicity to human skin fibroblasts compared to popular antibiotics. H2CBD performs similarly to CBD in terms of its properties and has the potential to be a valuable alternative.

Azolylpyrimidinediols as Novel Structural Scaffolds of DNA-Groove Binders against Intractable Acinetobacter baumannii

Summary: A unique class of antibacterial azolylpyrimidinediols (APDs) and their analogues were developed. Some synthesized compounds showed excellent bacteriostatic potency; especially, triazolylpyrimidinediol (triadol PD) 7a exhibited good anti Acinetobacter baumannii potential with a low MIC of 0.002 mmol/ L. Mechanistic explorations revealed that compound 7a without membrane-targeting ability could decrease metabolic activity, interact with DNA through groove binding action to block DNA replication rather than intercalate into and cleave DNA, and thus inhibit bacterial growth.

Structure-activity relationship studies of thiazole agents with potential anti methicillin-resistance Staphylococcus aureus (MRSA) activity

Summary: Due to the increasing resistance of MRSA to traditional antibiotics, there is a need for the development of new antimicrobial agents. Thiazoles conjugated with different heterocyclic ring systems are recognized as key frameworks for generating molecules with antimicrobial properties against MRSA. Molecular hybridization, which combines multiple heterocyclic pharmacophores into one molecule, offers a new approach for the development of novel agents with improved biological applications. This review focuses on recent advancements in thiazole-based hybrid molecules as anti-MRSA agents and discusses the structure-activity relationships (SARs) in different orientations.

Summary: The development of small molecule inhibitors as antibacterial agents against MRSA is a growing field in medicinal chemistry. Benzimidazoles, a class of heterocyclic compounds, have shown promising capability in fighting against MRSA due to their high biocompatibility, synergistic behavior with antibiotics, and tunable physicochemical properties. These compounds act as DNA binding agents, enzyme inhibitors, and anti-biofilm agents, showing a synergistic effect with available antibiotics to achieve antibacterial activity against MRSA. This review focuses on recent developments of benzimidazole-hybrid molecules as anti-MRSA agents, discussing their possible mechanism of action and structure-activity relationship.

Natural Berberine-Hybridized Benzimidazoles as Novel Unique Bactericides against Staphylococcus aureus

Mechanism studies indicated that the active molecule 7d damaged membrane integrity, stimulated ROS generation, and bound with DNA and S. aureus sortase A, exerting powerful antibacterial ability in treating S. aureus in the livestock industry.
