Recycling preparative isolation of six bicyclol active metabolites from SD rat urine using macroporous resin, offline 2D LPLC/HPLC, and prep-HPLC combined with pharmacodynamic evaluation of two active metabolites

Xucong Huang et al.

Summary: A highly efficient preparative approach was developed to extract six active metabolites of bicyclol (BIC) from rat urine. The prepared metabolites showed high purity and were verified by nuclear magnetic resonance (NMR). The evaluation of a liver injury zebrafish model revealed that one of the metabolites (M7) exhibited better effects than BIC in histopathology, gene expression, and aminotransferase levels, providing a strategy for discovering high-value candidate compounds from biological metabolism.

A robust, accurate, sensitive LC-MS/MS method to measure indoxyl sulfate, validated for plasma and kidney cells

Sabbir Ahmed et al.

Summary: Proximal tubular damage is a crucial prognostic factor in chronic kidney diseases, and indoxyl sulfate (IS) has the potential to be a biomarker for tubular function, as it accumulates in plasma during tubular dysfunction. A fast bioanalytical method using LC-MS/MS was developed and validated for measuring IS levels, showing promising results for its potential use as a biomarker for tubular damage in diabetic nephropathy.

Identification of bicyclol metabolites in rat plasma, urine and feces by UPLC-Q-TOF-MS/MS and evaluation of the efficacy and safety of these metabolites based on network pharmacology and molecular docking combined with toxicity prediction

Xucong Huang et al.

Summary: In this study, the metabolites of Bicyclol (BIC) were identified and their efficacy and safety were evaluated. The combination of network pharmacology and molecular docking analysis revealed that the metabolites had higher efficacy, better solubility, and lower toxicity compared to BIC. These findings contribute to a better understanding of the pharmacological mechanism of BIC and provide a basis for drug development.
