Update on the Discovery of Efflux Pump Inhibitors against Critical Priority Gram-Negative Bacteria

Nina Compagne et al.

Summary: Antimicrobial resistance (AMR) is a major public health problem that has caused an estimated 4.95 million deaths in 2019. The overuse of antibiotics has led to the development of bacterial strains that are resistant to known antibiotics. Multidrug efflux pumps, which are naturally expressed by Gram-negative bacteria, play a central role in AMR. This review focuses on eight families of efflux pump inhibitors (EPIs) that have shown activity against Escherichia coli or Pseudomonas aeruginosa, providing insights into their structure-activity relationships, synthesis, in vitro and in vivo activities, pharmacological properties, binding sites, and mechanisms of action.

Discovery of AcrAB-TolC pump inhibitors: Virtual screening and molecular dynamics simulation approach

Thien-Vy Phan et al.

Summary: This study aimed to establish a virtual screening model with prediction capabilities for efflux pump inhibitory activities. By analyzing candidate molecules from DrugBank and ZINC15 datasets with 2D and 3D models, three potential inhibitors were discovered. This research is of great significance for the discovery of new antibiotic therapies.

Identification of Phytoconstituents as Potent Inhibitors of Casein Kinase-1 Alpha Using Virtual Screening and Molecular Dynamics Simulations

Alaa Shafie et al.

Summary: In this study, three phytoconstituents, Semiglabrinol, Curcusone A, and Liriodenine, were identified as potential inhibitors of CK1 alpha with considerable affinity and specificity, stabilizing the protein and reducing conformational fluctuations. Molecular dynamics simulations and MM-PBSA analysis further confirmed the strong binding affinity of these compounds towards CK1 alpha.
