Inhibition of the C3a receptor attenuates sepsis-induced acute lung injury by suppressing pyroptosis of the pulmonary vascular endothelial cells

Zhan-Fei Li et al.

Summary: Acute lung injury (ALI) is a leading cause of bacterial sepsis-related death. Studies have shown that C3a levels are significantly increased in organs affected by sepsis and are closely associated with organ dysfunction and poor prognosis. This study found that C3a plays an important role in preventing sepsis-induced lung endothelial cell (EC) pyroptosis.

The suppressive effects of Mer inhibition on inflammatory responses in the pathogenesis of LPS-induced ALI/ARDS

Masahiko Fukatsu et al.

Summary: This study found that plasma concentrations of Gas6 and soluble Mer were higher in patients with severe sepsis or septic ALI/ARDS. The selective Mer inhibitor UNC2250 was shown to effectively inhibit LPS-induced lung injury in mouse models, suggesting its potential therapeutic use in treating inflammatory responses in ALI/ARDS.

LPA2 Contributes to Vascular Endothelium Homeostasis and Cardiac Remodeling After Myocardial Infarction

Jianqiu Pei et al.

Summary: This study found that the level of lysophosphatidic acid (LPA) was increased in the peripheral blood of mice after myocardial infarction (MI), and the expression of its receptor LPA(2) was also increased in the heart. Deficiency of Lpar2 led to impaired heart function, increased scar size, and reduced vascular density. Mechanistic studies demonstrated that LPA-LPA(2) signaling promoted angiogenesis and maintained vascular homeostasis through multiple signaling pathways.

The Two Sides of Siponimod: Evidence for Brain and Immune Mechanisms in Multiple Sclerosis

Stanley L. Cohan et al.

Summary: Siponimod is an approved oral treatment for relapsing forms of multiple sclerosis, which targets the immune response and neural inflammation through regulating S1P(1) and S1P(5) receptors. It reduces lymphocyte migration to the central nervous system and exerts effects on various brain cells to decrease neurodegeneration and damage.

The dipeptidyl peptidase-4 inhibitor linagliptin ameliorates LPS-induced acute lung injury by maintenance of pulmonary microvascular barrier via activating the Epac1/AKT pathway

Na Zhang et al.

Summary: This study found that linagliptin can effectively restore PMVECs barrier dysfunction and inhibit oxidative stress to protect against ALI by activating the Epac1/AKT signaling pathway, providing a potential new approach for clinical treatment of ALI.

Endothelial S1P1 Signaling Counteracts Infarct Expansion in Ischemic Stroke

Anja Nitzsche et al.

Summary: This study explores the role of endothelial cell S1P(1) in the naive and ischemic brain as a potential target for cerebrovascular therapy. Endothelial S1P(1) plays a critical vascular protective role in the mouse brain, sustaining endothelial functions and expanding during ischemia. Disruption of this pathway exacerbates brain injury in ischemic stroke models, highlighting the importance of S1P signaling in maintaining perfusion and microvascular patency in the ischemic penumbra.
