Antibiotic Adjuvants: A Versatile Approach to Combat Antibiotic Resistance

Geetika Dhanda et al.

Summary: The problem of antibiotic resistance is becoming more serious, with even the last resort antibiotics facing multidrug-resistant strains. The drug discovery process is often hindered by strict requirements for effective drug design. In such a situation, it is important to understand the different mechanisms of resistance and target them to improve antibiotic efficacy. Antibiotic adjuvants, nonantibiotic compounds that target bacterial resistance, can be used in combination with obsolete drugs for better therapeutic outcomes. This review discusses various acquired and inherent resistance mechanisms employed by bacteria and explores the use of antibiotic adjuvants to target these mechanisms. Different types of resistance breakers, such as enzyme inhibitors and efflux pump inhibitors, are discussed, as well as membrane-targeting compounds and host immune-modulating compounds. Challenges and potential directions for clinical translation of adjuvants are also addressed. Combinatorial therapy using antibiotic adjuvants has great potential as an alternative strategy to conventional antibiotic discovery.

Targeting Acyl Homoserine Lactones (AHLs) by the quorum quenching bacterial strains to control biofilm formation in Pseudomonas aeruginosa

Syeda Javariya Khalid et al.

Summary: Finding novel biological strategies to mitigate bacterial biofilms is crucial for combating bacterial infections as biofilm-forming bacteria are highly resistant to antibiotics. This study investigated the potential of quorum quenching (QQ) enzymes, which interfere with the quorum sensing (QS) mechanism, to mitigate biofilms. Bacterial strains with QQ activity were identified and tested against multidrug-resistant Pseudomonas aeruginosa. The results showed that certain QQ strains were able to reduce biofilm formation by up to 60%. The presence of AHL-degrading enzymes was confirmed, providing insights into the mechanism of biofilm mitigation.

Microbiologically-Synthesized Nanoparticles and Their Role in Silencing the Biofilm Signaling Cascade

Dibyajit Lahiri et al.

Summary: The rise of bacterial resistance to antibiotics has led to the exploration of alternative antimicrobial treatment strategies, with engineered nanoparticles becoming more common for efficient penetration into living systems. Microbial enzymes/proteins are increasingly being used as reducing agents for synthesizing nanoparticles due to their speed, environmental safety, and cost-effectiveness. Fungi and bacteria are preferred biogenic sources for their ability to produce high levels of reductase enzymes and regulate the size and morphology of synthesized nanoparticles, which can reduce manufacturing costs. The penetration of nanoparticles through biofilm matrix allows them to inhibit bacterial growth and disrupt cell-to-cell communication mechanisms, ultimately preventing biofilm formation and associated chronic diseases.
