ROS/PI3K/Akt and Wnt/β-catenin signalings activate HIF-1α-induced metabolic reprogramming to impart 5-fluorouracil resistance in colorectal cancer

Shuohui Dong et al.

Summary: This study describes the abnormality of glucose metabolism in 5-FU-resistant CRC, and shows that enhanced glycolysis and pentose phosphate pathway are associated with increased HIF-1 alpha expression. HIF-1 alpha-induced glucose metabolic reprogramming imparts 5-FU resistance in CRC. HIF-1 alpha is a potential biomarker for 5-FU-resistant CRC, and targeting HIF-1 alpha in combination with 5-FU may represent an effective therapeutic strategy in 5-FU-resistant CRC.

Papillary thyroid carcinoma tall cell variant shares accumulation of mitochondria, mitochondrial DNA mutations, and loss of oxidative phosphorylation complex I integrity with oncocytic tumors

Oleksiy Tsybrovskyy et al.

Summary: The study identified similarities between papillary thyroid carcinoma tall cell variant (PTC-TCV) and oncocytic thyroid tumors, including mitochondrial accumulation and loss of oxidative phosphorylation (OXPHOS) integrity. MtDNA mutations in PTC-TCV exhibited high heteroplasmy, while these mutations were less common in cPTC.

Current status of liver transplantation for cholangiocarcinoma

Patrick Twohig et al.

Summary: Cholangiocarcinoma is the second most common liver cancer with a relatively short median survival without treatment. Surgical resection is the main treatment method, but a large percentage of tumors are inoperable. However, improvements in patient selection criteria and neoadjuvant treatment protocols have shown promise in improving outcomes for inoperable patients.

The Role of Mitochondria in the Chemoresistance of Pancreatic Cancer Cells

Yibo Fu et al.

Summary: Mitochondria play important roles in the chemoresistance of pancreatic cancer by affecting apoptosis, metabolism, mtDNA metabolism, and mitochondrial dynamics. Targeting certain factors in mitochondria may improve the sensitivity of pancreatic cancer cells to chemotherapeutic agents.

Cell Death and Inflammation: The Role of Mitochondria in Health and Disease

Anna Picca et al.

Summary: Mitochondria play a central role in cellular processes and maintaining organelle homeostasis is crucial for efficient mitochondrial function. Mechanisms of mitochondrial quality control, such as dynamics, biogenesis, proteostasis, and autophagy, safeguard organelle integrity and function. The crosstalk between autophagy and apoptosis plays a key role in regulating mtDNA release, cell fate, and inflammation. Interferon production during mtDNA-mediated inflammation may be utilized for the elimination of dying cells and promoting anti-tumor immunity.

Cullin3 deficiency shapes tumor microenvironment and promotes cholangiocarcinoma in liver-specific Smad4/Pten mutant mice

Ming Zhao et al.

Summary: The study identified 15 potential tumor suppressor genes including Cullin3 (Cul3), which when deficient accelerates cholangiocytes expansion leading to the initiation of cholangiocarcinoma (CC). Cul3 deficiency also induces inflammation and promotes the progression of CC.
