Recent advances in biological production of 1,3-propanediol: new routes and engineering strategies

Systems metabolic engineering of Corynebacterium glutamicum for high-level production of 1,3-propanediol from glucose and xylose

Summary: In this study, an efficient C. glutamicum strain for the production of 1,3-propanediol (1,3-PDO) from glucose and xylose was developed using systems metabolic engineering approaches. Through the construction and optimization of glycerol and 1,3-PDO synthesis modules, as well as pathway engineering to reduce toxic byproducts, the engineered strain demonstrated high production of 1,3-PDO. This study highlights the potential of C. glutamicum as a chassis for the industrial production of 1,3-PDO from lignocellulosic feedstocks.

Modification of substrate and product transport systems in Klebsiella pneumoniae to improve 1,3-propanediol production

Summary: This study focused on promoting 1,3-propanediol (1,3-PDO) production in Klebsiella pneumoniae by overexpressing the glycerol uptake facilitator (GlpF) and resistance-nodulation-cell division (RND) type efflux system members. Overexpression of GlpF improved glycerol dehydratase (GDHt) activity and increased 1,3-PDO production. Different members of the RND system had varying effects on 1,3-PDO production, with AcrA and AcrE showing no impact and AcrF and TolC increasing 1,3-PDO titer. MexB significantly decreased GDHt activity and 1,3-PDO production, while MexF dramatically enhanced both GDHt activity and 1,3-PDO production.

Summary: The disruption of the glpF gene in engineered Escherichia coli increases the intracellular concentration of glycerol, leading to higher 1,3-propanediol (1,3-PDO) production. This disruption also results in a decrease in the by-product acetic acid and enhances the specific growth rate and 1,3-PDO yield from glucose compared to the non-disrupted strain.
