Tight junction proteins related to blood-brain barrier and their regulatory signaling pathways in ischemic stroke

Abnormal expression of TJ-related proteins can lead to TJ destruction, structural damage, and dysfunction of endothelial and epithelial cells, which are associated with the occurrence and progression of various diseases. The blood-brain barrier (BBB) damage and dysfunction are significant pathological features of stroke, and TJs have a direct impact on BBB integrity. This article discusses the structure, function, regulatory mechanisms, and therapeutic potential of BBB TJs in ischemic stroke.

This study isolated normal brain pericytes (nPCs) and compared them with ischemic pericytes (iPCs) in a mouse model of ischemic stroke. Summary: This study evaluated the neuroprotective effects of nmFGF1 on microglia in a mouse stroke model and determined its mechanism of action. The findings showed that nmFGF1 reduced infarct size and improved neurological deficits by regulating the secretion of inflammatory and anti-inflammatory cytokines. In vitro experiments demonstrated that nmFGF1 modulated the expression of these cytokines under oxygen-glucose deprivation and lipopolysaccharide stimulation. Furthermore, nmFGF1 activated the Nrf2 pathway and inhibited the NF-kappa B pathway, leading to a reduction in neuroinflammation. These results suggest that nmFGF1 has the potential to be used as a therapeutic agent for ischemic stroke.
