Bayesian optimal phase II designs with dual-criterion decision making

Summary: The conventional phase II trial design paradigm based on hypothesis testing may not provide sufficient evidence of clinical effectiveness. This study proposes the Bayesian optimal phase II trial design with dual-criterion decision making (BOP2-DC) that incorporates statistical significance and clinical relevance. BOP2-DC allows for go/consider/no-go decisions based on the posterior probabilities of statistical and clinical significance. The design is flexible and can accommodate various types of endpoints in both single-arm and randomized trials.
