AGGF1 therapy inhibits thoracic aortic aneurysms by enhancing integrin α7-mediated inhibition of TGF-β1 maturation and ERK1/2 signaling

Summary: Thoracic aortic aneurysm (TAA) causes numerous sudden deaths annually, yet there is no effective pharmacologic intervention. This study demonstrates that AGGF1 can effectively inhibit TAA-associated arterial inflammation and remodeling in three different mouse models. AGGF1 expression is decreased in the ascending aortas of these models as well as in human TAA patients. Pirfenidone, a drug used for pulmonary fibrosis, also shows potential in inhibiting TAA-associated vascular inflammation and remodeling. Therefore, AGGF1 and TGF-β inhibition therapies hold promise for treating TAA.

In utero delivery of miRNA induces epigenetic alterations and corrects pulmonary pathology in congenital diaphragmatic hernia

Summary: Using a rat model of CDH, this study investigated the expression of miR200b and the TGF-beta pathway at different gestational times. The results showed that delivering miR200b-loaded nanoparticles to fetal rats with CDH via vitelline vein injection resulted in changes in the TGF-b pathway, leading to improved lung size, morphology, and pulmonary vascular remodeling. This is the first demonstration of in utero epigenetic therapy to improve lung growth and development.

Ginsenoside Rg1 ameliorates hypoxia-induced pulmonary arterial hypertension by inhibiting endothelial-to-mesenchymal transition and inflammation by regulating CCN1

Summary: The aim of this study was to investigate the therapeutic effect of ginsenoside Rg1 on hypoxia-induced pulmonary arterial hypertension (HPH). The results showed that ginsenoside Rg1 could reverse hypoxia-induced endothelial-mesenchymal transition (EndMT) and inflammation by regulating CCN1, which has potential value in the prevention and treatment of HPH. This study revealed the role of EndMT and inflammation in HPH and identified a potential treatment option.

Summary: This study provides evidence that vioprolide A (vioA) is an interesting compound with anti-angiogenic and anti-inflammatory effects. VioA reduced angiogenic features in endothelial cells by decreasing VEGFR2 protein levels and TAZ signaling. Surprisingly, vioA also promoted cell survival by activating Akt and inhibiting apoptosis. Further preclinical studies on vioA are warranted based on these findings.

Imbalance between relaxing and contracting factors derived from the endothelium leads to endothelial dysfunction, which is a critical contributor in aging and chronic cardiometabolic disorders. Oxidative stress and inflammation are key players in endothelial dysfunction and are considered as potential targets for intervention in patients with cardiovascular and metabolic diseases.
