Promising antivirals for PLpro of SARS-CoV-2 using virtual screening, molecular docking, dynamics, and MMPBSA

Summary: This study screened a set of antiviral molecules and identified five molecules with good binding affinities to the papain-like protease (PLpro) of SARS-CoV-2. Molecular dynamics analysis and Molecular Mechanics/Position-Boltzmann Surface Area (MMPBSA) analysis confirmed the stability and lower energy of the PLpro-inhibitor complexes. The results provide promising candidates for potential antiviral drugs against SARS-CoV-2.

Insights into the enhanced thermal stability of lysozyme with altered structure and activity induced by choline chloride-based deep eutectic solvents containing polyols and sugars

Summary: This study systematically investigated the effect of deep eutectic solvents (DESs) on the stability, structure, and activity of proteins using lysozyme as a model. The results showed that DESs can improve the thermal stability and alpha-helical content of lysozyme, indicating a protective effect. The impact of DESs on lysozyme varies with different concentrations, and DESs with polyol as the hydrogen bond donor exhibit better stability and preservation of the native structure compared to those with sugar as the donor.

Assessing the Influence of Betaine-Based Natural Deep Eutectic Systems on Horseradish Peroxidase

Summary: This study validates the use of horseradish peroxidase (HRP) in natural deep eutectic systems (NADES). The water activity of the NADES was found to be around 0.4 at 37 degrees C, with water contents ranging from 14% to 22%. The density and viscosity of the NADES were determined experimentally and through thermodynamic modeling. The study also assessed the impact of the NADES on enzymatic activity and the conformational and thermal stability.

Assessing the Influence of Betaine-Based Natural Deep Eutectic on Horseradish Peroxidase

Structure-Based Identification of Potential Drugs Against FmtA of Staphylococcus aureus: Virtual Screening, Molecular Dynamics, MM-GBSA, and QM/MM

Summary: Staphylococcus aureus is resistant to beta-lactam antibiotics due to the presence of FmtA, an esterase that plays a key role in methicillin resistance. Computational drug screening identified ofloxacin, roflumilast, and furazolidone as potential molecules with high binding affinities to FmtA. Molecular dynamics simulations and analysis revealed that the FmtA-drug complexes are stable and the residues in the active site of FmtA are essential for forming stable protein-ligand complexes with lower energy. Further testing and modification of these screened molecules are needed to develop antimicrobial compounds against S. aureus.

Effects of processing conditions on hydrolysates of proteins from whole whey and formation of Maillard reaction products

Summary: Chemical modifications of proteins during food processing can lead to new functionalities and applications of products. This study demonstrates that pH has the strongest effect on the release of peptides during whey protein hydrolysis, and significantly affects the generation of MRPs and ferric reducing capacity of the hydrolysates. The hydrolytic processes could enhance the antioxidant capacity of whey for use as natural preservatives in the food industry.

Deep eutectic systems from betaine and polyols - Physicochemical and toxicological properties

Summary: The tailor-made versatility of deep eutectic systems (DES) has opened promising future perspectives for their applications in industry, with easily tunable properties. Results showed that the DES tested generally had low toxicity profile, which could be substantially reduced by increasing their water content. This suggests that the studied DES can be minimally toxic and have potential for use in green technologies.
