Novel application of rhein and its prodrug diacerein for reversing cancer-related multidrug resistance through the dual inhibition of P-glycoprotein efflux and STAT3-mediated P-glycoprotein expression

Summary: This study evaluated the P-gp inhibiting and MDR-reversing effects of two compounds, rhein and diacerein, in cancer cells. The results showed that both compounds can inhibit P-gp efflux function and downregulate P-gp expression by interacting with STAT3. In addition, rhein demonstrated a strong MDR-reversing effect, while diacerein showed selective cytotoxicity and mild MDR-reversing ability.

Sesquiterpene Lactones Attenuate Paclitaxel Resistance Via Inhibiting MALAT1/STAT3/FUT4 Axis and P-Glycoprotein Transporters in Lung Cancer Cells

Summary: This study reveals a positive correlation between the non-coding RNA MALAT1 and the activities of STAT3 and FUT4 in paclitaxel resistant lung cancer cells. Two natural compounds, alantolactone (ALT) and Brevilin A (Brv-A), induce apoptosis in these cells and suppress the expression of MALAT1, STAT3, FUT4, and P-GP, which are associated with paclitaxel resistance. Inhibition of MALAT1 enhances the effectiveness of ALT and Brv-A, leading to growth inhibition, as well as anti-migratory and anti-invasive effects.

Summary: Arctigenin is a bioactive compound found in Chinese herbal medicines that has shown anti-inflammatory, anti-tumor, and immunomodulatory properties. It can induce apoptosis in tumor cells through multiple pathways, inhibit inflammation in the tumor microenvironment, and downregulate factors related to tumor metastasis and angiogenesis. In addition, Arctigenin can abolish tumor cell growth in nutrient-deprived conditions by modulating MAPK signaling pathways. Further research is being done to enhance its solubility and pharmacogenetic properties for potential use in future chemotherapeutic approaches.
