EGFR-targeted humanized single chain antibody fragment functionalized silica nanoparticles for precision therapy of cancer

Analysis of the in vitro function and internalization ability of a humanized EGFR antibody AE01 expressed by Chinese hamster ovary cells

Summary: The objective of this study was to produce humanized EGFR antibody and investigate its in vitro binding and internalization to EGFR-overexpressing A431 epidermoid carcinoma cells. Firstly, humanized anti-EGFR AE01 was stably expressed in CHO system. The expression of AE01 was confirmed by SDS-PAGE and Western blot. The binding and internalization of AE01 were examined using flow cytometry and immunofluorescence assay. The results demonstrated that: (1) Pure humanized AE01 was successfully prepared, (2) AE01 specifically bound to EGFR-positive A431 cells on the cell surface, but not to EGFR-negative NIH 3T3 cells, (3) AE01 effectively inhibited the proliferation of A431 cells, and (4) AE01 binding to A431 cell surface triggered internalization. The antibody could potentially serve as a candidate molecule for targeted therapeutic vectors in EGFR overexpressed cancer cells.
