A novel power prior approach for borrowing historical control data in clinical trials

Yaru Shi et al.

Summary: There is growing interest in using historical control data to improve statistical power and reduce sample sizes in clinical trials. However, accounting for heterogeneity between historical and current trials can be challenging. This paper proposes a novel approach to selecting a power prior based on direct measures of distributional differences between historical and current control data. Simulations show that the proposed power prior improves study power and controls type I errors when the distribution of historical control data is similar to current control data.

New approaches for testing non-inferiority for three-arm trials with Poisson distributed outcomes

Samiran Ghosh et al.

Summary: With limited resources, it is crucial to innovate statistical methods for the design and analysis of randomized controlled trials (RCTs) in order to discover newer and better treatments. Non-inferiority (NI) testing allows for less efficacious treatment options that may be favorable for certain subgroups of patients. This article proposes novel Frequentist and Bayesian procedures for testing NI in three-arm trials with Poisson distributed count outcome, which is common in various disease areas. The Bayesian approach is particularly advantageous when historical information is available. Sample size calculation and the connection between the two paradigms are also discussed.

Bayesian sample size determination in a three-arm non-inferiority trial with binary endpoints

Niansheng Tang et al.

Summary: This article introduces a design and analysis method for three-arm non-inferiority trials, and proposes two fully Bayesian approaches to determine the required sample size. Simulation studies validate the performance of the proposed methods, and an example is provided to demonstrate their application.

A novel equivalence probability weighted power prior for using historical control data in an adaptive clinical trial design: A comparison to standard methods

Summary: The study introduces two novel approaches for incorporating historical control data in trial design, improving efficiency and reducing required controls, but there is a need to consider consistency between historical and current data to avoid bias.

COVID-19 vaccine trials: The use of active controls and non-inferiority studies

Summary: Non-inferiority trials aim to evaluate the efficacy of experimental vaccines by comparing them to established effective active control vaccines. This article explores non-inferiority margins to address global vaccine needs and proposes methods to reliably assess experimental vaccine efficacy.
