Design, synthesis and antitumor evaluation of novel pyrazolo[3,4-d]pyrimidines incorporating different amino acid conjugates as potential DHFR inhibitors

Summary: This study investigated the antitumor effect of simultaneous inhibition of dihydrofolate reductase (DHFR) enzyme. Novel pyrazolo[3,4-d]pyrimidines were designed as efficient antifolate agents, similar in structure to methotrexate (MTX) and MTX-related antifolates. Compound 7f demonstrated the most potent antitumor activity by arresting cell cycle and inducing apoptosis.

Summary: A series of coumarin derivatives were designed, synthesized, and evaluated for their antiproliferative activity. Compound 3e exhibited significant antiproliferative activity and inhibited VEGFR-2 with an IC50 value of 0.082 +/- 0.004 mu M. It also caused cell cycle arrest at the G1/S phase. Compound 3b showed the most active antibacterial activity against K. pneumonia, with the same MIC and MBC value of 128 mu g/mL.

Compounds 6b, 6f, and 6g showed potent anticancer activity at 10 mu M dose. Compound 6f exhibited broad-spectrum antiproliferative activity, inhibited VEGFR-2, reduced angiogenesis, induced apoptosis, and arrested the cell cycle at the S phase in HCT-15 colon cancer cells.

Molecular modeling studies were conducted to understand the binding pattern of the active compounds.

Summary: A series of 20 thieno[2,3-d]pyrimidine derivatives were synthesized and evaluated for their antiproliferative activity against 60 cancer cell lines. Compound 7b showed significant antineoplastic activity and inhibited the growth of 37 tested cancer cell lines at micromolar concentrations. It effectively inhibited EGFR and showed downregulation effects on total EGFR concentration and its phosphorylation. Compound 7b also increased apoptosis and arrested the cell cycle in the G1/S phase.

Design, synthesis and evaluation of anticancer activity of new pyrazoline derivatives by down-regulation of VEGF: Molecular docking and apoptosis inducing activity

Summary: Two series of pyrazoline compounds were synthesized as antiproliferative agents targeting the VEGFR pathway. Compound 3f showed the highest anticancer activity against ovarian and breast cancer cell lines, inhibiting cell proliferation and inducing apoptosis. It also down-regulated the expression of VEGF and p-VEGFR-2. Docking studies revealed its similar interaction pattern to axitinib on VEGFR-2 receptor and its ability to fit into the STAT3 binding site, explaining the down-regulation of VEGF.

These findings suggest that 7f has the potential to be a targeted therapeutic drug for breast cancer and leukemia.

Summary: In this study, a series of novel benzoxazole derivatives were designed, synthesized, and evaluated for their potential as VEGFR-2 inhibitors. Compound 12l showed high growth inhibitory activity against HepG2 and MCF-7 cell lines and demonstrated promising VEGFR-2 inhibitory activity. Further investigation revealed that 12l could arrest cell growth at the Pre-G1 and G1 phases, induce apoptosis, and regulate key proteins involved in apoptosis. Docking studies suggested that 12l interacted with key amino acids similar to sorafenib.

Summary: A novel series of pyrazolo[3,4-d]pyrimidine derivatives bearing carbon-aryl(heteryl)idene moieties was synthesized, with compound 10k showing potent anticancer activity by inducing G2/M phase arrest and decreasing mitochondrial membrane potential. It also exhibited significant inhibition of cell migration and tube formation in HUVEC. Molecular modeling studies indicated that 10k may exert its anticancer effects by forming hydrogen bonds with VEGFR-2 and inhibiting tubulin polymerization.
