Microspore embryogenesis induction by mannitol and TSA results in a complex regulation of epigenetic dynamics and gene expression in bread wheat

Isabel Valero-Rubira et al.

Summary: Reprogramming microspores development towards embryogenesis through stress treatment is crucial for doubled haploid production. This study investigates the epigenetic dynamics and expression of genes associated with histone post-translational modifications (PTMs) and microspore embryogenesis (ME) induction in bread wheat cultivars. The results reveal that the induction efficiency of ME is related to the epigenetic state and expression of genes involved in pollen development, auxin signaling, and hormone signaling pathways. This breakthrough study provides insights into the molecular mechanisms governing ME induction efficiency in recalcitrant wheat genotypes and other crops.

Effects of TSA, NaB, Aza in Lactuca sativa L. protoplasts and effect of TSA in Nicotiana benthamiana protoplasts on cell division and callus formation

Seung Hee Choi et al.

Summary: Whole-plant regeneration through plant tissue culture is a complex process regulated by genetic and environmental factors. Epigenetic regulation has been found to play a crucial role in plant cell differentiation and pluripotency. In this study, the effects of epigenetic regulators on lettuce and tobacco protoplasts were tested, and it was found that these chemicals promoted cell division and callus formation. These findings suggest that these chemicals can be used to enhance plant regeneration and crop improvement.

The genetic diversity of Ethiopian barley genotypes in relation to their geographical origin

Surafel Shibru Teklemariam et al.

Summary: The genetic diversity of Ethiopian barley, influenced by diverse topography, altitude, climate conditions, soil types, and farming systems, was analyzed. Three genetically distinct clusters were identified, with higher genetic variation within clusters and moderate genetic differentiation between clusters. The weak correlation between geographic distance and genetic differentiation was observed, but certain regions showed a significant association between landraces and genetic clusters.

Insights into the Histone Acetylation-Mediated Regulation of the Transcription Factor Genes That Control the Embryogenic Transition in the Somatic Cells of Arabidopsis

Joanna Moronczyk et al.

Summary: Somatic embryogenesis (SE) is a complex process involving the reprogramming of plant somatic cells, and histone acetylation plays a crucial role in regulating the expression of key transcription factor genes during SE. This study investigated the global and gene-specific changes in histone acetylation in Arabidopsis explants undergoing SE and identified H3 and H4 acetylation as potential regulators of critical TF genes. The findings also revealed that HDA19 negatively affects SE by regulating LEC1, LEC2, and BBM, and provided evidence of HDA19's involvement in histone acetylation-mediated regulation of LEC2 during SE.

Induction of Somatic Embryogenesis in Tamarillo (Solanum betaceum Cav.) Involves Increases in the Endogenous Auxin Indole-3-Acetic Acid

Andre Caeiro et al.

Summary: Somatic embryogenesis in tamarillo is a complex process regulated by various factors, including the plant growth regulator indole-3-acetic acid (IAA). This study investigated the endogenous levels of IAA during induction and in calli samples using chemical quantification and immunohistochemistry. The results showed that embryogenic calli had higher levels of IAA compared to non-embryogenic calli, and the degradation rates of IAA were related to these levels. Immunohistochemical analysis supported these findings, and differential gene expression analysis revealed distinct molecular responses between embryogenic and non-embryogenic calli.

Identification of regulatory factors promoting embryogenic callus formation in barley through transcriptome analysis

Jingqi Suo et al.

Summary: This study identified different transcription regulation pathways between calli formed from immature and mature embryos in barley, revealing significant changes in gene expression profiles induced by auxin-rich medium within 48 hours. The differential expression of IAA genes suggests the involvement of auxin response in regulating cell reprogramming during callus formation. Additionally, epigenetic regulators and key genes involved in meristem development were found to play crucial roles in determining the acquisition of competency, contributing to improved transformation efficiency in barley.

Summary: De novo shoot organogenesis (DNSO) is a common procedure used for shoot regeneration in vitro. The genetic regulation of DNSO has been studied in detail, highlighting the roles of plant hormones cytokinin (CK) and auxin, as well as their interaction with sucrose in the regeneration media. These signaling events mediate the complex process of shoot regeneration.

Histone Deacetylase Inhibitors Increase the Embryogenic Potential and Alter the Expression of Embryogenesis-Related and HDAC-Encoding Genes in Grapevine (Vitis vinifera L., cv. Mencia)

Oscar Martinez et al.

Summary: The use of an induction medium containing histone deacetylase inhibitors, like sodium butyrate, improved the embryogenic responses in grapevine cotyledonary and recently germinated somatic embryos. The study also showed significant overexpression of certain grapevine genes related to embryogenic competence, indicating a potential molecular response triggered by the inhibitor for enhancing embryogenic competence. This treatment also led to significant variations in the expression of histone deacetylase enzyme-encoding genes, offering possibilities for obtaining somatic embryos and reducing seasonal constraints in grapevine cultivation.
