Identification of potent inhibitors for Leishmania donovani homoserine kinase: an integrated in silico and kinetic study

Discovery of Natural Bisbenzylisoquinoline Analogs from the Library of Thai Traditional Plants as SARS-CoV-2 3CLPro Inhibitors: In Silico Molecular Docking, Molecular Dynamics, and In Vitro Enzymatic Activity

Summary: We used computer-assisted drug screening to investigate 326 natural products from Thai traditional plants for their potential as anti-SARS-CoV-2 drugs. After virtual screening, we identified seven bisbenzylisoquinolines that showed higher binding affinity and stability than existing inhibitors. In vitro enzymatic assays confirmed their inhibitory activity against SARS-CoV-2 3CLPro, with isoliensinine (5) showing the highest inhibition. These findings suggest that bisbenzylisoquinoline scaffolds could be used for the development of effective anti-SARS-CoV-2 drugs.

Development of novel dipeptide nitriles as inhibitors of rhodesain of Trypanosoma brucei rhodesiense

Summary: In this study, a new series of dipeptide nitriles were developed as reversible rhodesain inhibitors at a nanomolar level. These compounds were shown to directly bind to the active site of rhodesain and act as competitive inhibitors. The most promising compounds exhibited high binding affinity and good antiparasitic activity, making them potential lead compounds for the discovery of new drugs to treat Human African Trypanosomiasis.

GROMACS in the Cloud: A Global Supercomputer to Speed Up Alchemical Drug Design
