The Cryptococcus neoformans Flc1 Homologue Controls Calcium Homeostasis and Confers Fungal Pathogenicity in the Infected Hosts

Summary: This study reveals the importance of the Flc1 protein in maintaining calcium homeostasis and vacuolar biogenesis in Cryptococcus neoformans. Deletion of Flc1 leads to an increase in cytosolic calcium concentration, an elevation of Crz1 transcription factor in the nucleus, and an aberrant accumulation of cell wall chitin. Flc1 is also involved in vacuolar fusion and capsule formation. The absence of Flc1 results in the loss of survival ability and virulence in Cryptococcus.

Summary: The mechanism of Ca2+-dependent regulation of TRPY1 and possible contributions of Ca2+-binding proteins are not well understood. The study shows a Ca2+-dependent binding of yeast calmodulin (CaM) to TRPY1. Furthermore, mammalian CaM can replace yeast CaM for inhibitory control of TRPY1.

A Transient Receptor Potential-like Calcium Ion Channel in the Filamentous Fungus Aspergillus nidulans

Summary: The putative TRP-like calcium channel trpR in the filamentous fungus Aspergillus nidulans plays important roles in conidiation and adapting to cell wall disruption reagents, especially under calcium-limited conditions. TrpR has an opposite response to the previously identified high-affinity calcium channels CchA/MidA in reacting to cell wall disruption reagents and regulating calcium transients. However, an addition of calcium can rescue the defects in TrpR and CchA/MidA, indicating calcium's ability to bypass the necessary requirement.
