In silico designing of an epitope-based peptide vaccine cocktail against Nipah virus: an Indian population-based epidemiological study

In-Silico Vaccine Design Based on a Novel Vaccine Candidate Against Infections Caused by Acinetobacter baumannii

Summary: This study focuses on developing a rational vaccine design against Acinetobacter baumannii using epitope mapping technique. By selecting an outer membrane protein with immunogenic potential, epitopes were calculated for selection based on certain criteria. A vaccine sequence of 240 amino acids was obtained by joining all the epitopes in a sequence-wise manner and adding a protein agonist to human innate immune receptors. The designed construct showed enhanced overall protein stability, expression, immunostimulatory capabilities, and solubility. Molecular dynamics simulations and molecular docking demonstrated the compactness, stability, and strong binding of the vaccine with Toll-like receptors. In-silico immune simulations indicated a significant increase in T-cell and B-cell populations. Bioinformatic tools were also useful in optimizing codons for successful cloning of constructs into host vectors.

In Silico Studies of Tumor Targeted Peptide-Conjugated Natural Products for Targeting Over-Expressed Receptors in Breast Cancer Cells Using Molecular Docking, Molecular Dynamics and MMGBSA Calculations

Summary: In this study, in silico studies were conducted to design diterpene and polyphenol-peptide conjugates that can potentially target over-expressed breast tumor cell receptors. The mutated peptides were created by inducing point mutations into a known tumor-targeting peptide sequence. Molecular docking studies and molecular dynamics simulations were performed to study the binding interactions and stability of the peptide conjugates with the receptors. The results showed enhanced binding interactions and stable receptor-ligand complexes, suggesting the potential therapeutic applications of these conjugates.

Summary: Since the beginning of the COVID-19 pandemic, various variants of SARS-CoV-2 have emerged, including five variants of concern. These variants, especially Omicron, have been shown to evade the immune response induced by existing COVID-19 vaccines to varying degrees, posing a significant risk to global health. In this study, a vaccine based on inactivated SARS-CoV-2 and an adjuvant was developed and tested in rats. The candidate vaccine elicited high levels of neutralizing antibodies against not only the original SARS-CoV-2, but also the Beta, Delta, and Omicron variants.

An Immunoinformatics Prediction of Novel Multi-Epitope Vaccines Candidate Against Surface Antigens of Nipah Virus
