TC2N Promotes Cell Proliferation and Metastasis in Hepatocellular Carcinoma by Targeting the Wnt/I3-Catenin Signaling Pathway

N6-Methyladenosine-Mediated Up-Regulation of FZD10 Regulates Liver Cancer Stem Cells' Properties and Lenvatinib Resistance Through WNT/b-Catenin and Hippo Signaling Pathways

Summary: This study reveals that the expression of Frizzled-10 (FZD10) in liver cancer stem cells is regulated by METTL3-mediated N6-methyladenosine methylation of FZD10 mRNA, and FZD10 promotes self-renewal, tumorigenicity, and metastasis of liver CSCs via activating β-catenin and YAP1. The FZD10-β-catenin/YAP1 axis is activated in liver CSCs and indicates poor prognosis. Furthermore, the FZD10/β-catenin/c-Jun/MEK/ERK axis determines the responses of hepatoma cells to lenvatinib treatment.

Summary: This study finds that G alpha i2 can induce the condensation of conductin, leading to the degradation of β-catenin and inhibition of Wnt signaling, thus suppressing the growth of colorectal cancer. Colorectal cancer evades this inhibition by decreasing the expression of G alpha i2 and inactivating mutations, but the G alpha i2-activating drug guanabenz can reduce colorectal cancer growth.

It perturbs CTNNB1 translocation and interaction with transcription factors independently of GSK3beta. The activation of DOC2B-DKK1-senescence-Wnt/beta-catenin-EMT signaling crosstalk may provide a novel approach to restrain malignant behaviors in cervical cancer.
