Understanding the signaling cascades in CAFs helps us better understand their roles in cancer progression and may lead to the development of more effective and safer therapies.

Summary: This Review summarizes the current understanding of lipid droplet biogenesis and functions in health and disease, emphasizing the crucial role of lipid droplet biogenesis in alleviating cellular stress. Lipid droplets are intracellular organelles that play diverse physiological and pathological roles beyond energy storage. Although progress has been made in understanding their biogenesis and functions, there are still gaps in our knowledge. Furthermore, the causal relationship between lipid droplet biogenesis and human diseases is not well established. This Review also discusses potential therapeutic strategies targeting lipid droplet biogenesis, growth, or degradation for common diseases such as cancer, hepatic steatosis, and viral infection.

Peritoneal high-fat environment promotes peritoneal metastasis of gastric cancer cells through activation of NSUN2-mediated ORAI2 m5C modification

Summary: Peritoneal metastasis (PM) is a significant mode of metastasis in gastric cancer (GC), associated with poor prognosis. However, the molecular mechanism of PM remains unclear. This study revealed that NSUN2, an RNA modifier, is upregulated in PM and patients with high NSUN2 expression have a worse prognosis. Mechanistically, NSUN2 regulates ORAI2 mRNA stability through m5C modification, promoting ORAI2 expression and resulting in peritoneal metastasis and colonization in GC.

Intraperitoneal metastasis of ovarian cancer: new insights on resident macrophages in the peritoneal cavity

Summary: Ovarian cancer metastasis primarily occurs in the peritoneal cavity, where the interaction between cancer cells and various cell types, particularly macrophages, creates a favorable environment for metastasis. In recent years, the heterogeneities of macrophages in different organs and their roles in tumor settings have been studied extensively. This review focuses on the unique microenvironment of the peritoneal cavity, including the peritoneal fluid, peritoneum, and omentum, as well as the resident macrophage populations. It summarizes the contributions of resident macrophages in ovarian cancer metastasis and discusses potential therapeutic strategies targeting these cells.

Summary: This study describes the abnormality of glucose metabolism in 5-FU-resistant CRC, and shows that enhanced glycolysis and pentose phosphate pathway are associated with increased HIF-1 alpha expression. HIF-1 alpha-induced glucose metabolic reprogramming imparts 5-FU resistance in CRC. HIF-1 alpha is a potential biomarker for 5-FU-resistant CRC, and targeting HIF-1 alpha in combination with 5-FU may represent an effective therapeutic strategy in 5-FU-resistant CRC.
