PGRMC1: An enigmatic heme-binding protein

Meredith R. McGuire et al.

Summary: Progesterone Receptor Membrane Component 1 (PGRMC1) is a heme-binding protein involved in various cellular and tissue functions. However, the underlying mechanisms of PGRMC1 in these biological aspects remain largely unknown. This review provides an overview of the literature on PGRMC1, highlighting supported information and areas for further investigation. The central role of PGRMC1 in cytochrome P450 activity is extensively discussed, and a speculative model is proposed regarding its function as a heme chaperone. By identifying knowledge gaps, this review aims to inspire a better understanding of this enigmatic protein.

A Comprehensive Evaluation of the Effects of RNA-Editing Proteins ADAR and ADARB1 on the Expression of the Drug-Metabolizing Enzymes in HepaRG Cells

Joseph M. Collins et al.

Summary: This study systematically surveyed the roles of ADAR and ADARB1 in both basal and induced expression of drug-metabolizing enzymes and assessed their coexpression in liver samples. The results support that ADAR and ADARB1 regulate the expression of drug-metabolizing enzymes in the liver, suggesting that factors affecting ADAR expression also have the potential to impact drug metabolism.

Summary: This study aimed to determine the genetic variants associated with hyperbilirubinemia and elevated ALT and AST levels in children, adolescents, and young adults treated for ALL. The results showed that UGT1A1 variant rs887829 and PNPLA3 variant rs738409 were associated with an increased risk of hyperbilirubinemia and elevated ALT and AST levels during ALL treatment.

Progesterone receptor membrane component 1 (PGRMC1) binds and stabilizes cytochromes P450 through a heme-independent mechanism

Meredith R. McGuire et al.

Summary: PGRMC1, a heme-binding protein, independently stabilizes a wide range of cytochromes P450 and supports their maintenance at the protein level. This study reveals a crucial heme-independent function of PGRMC1 in cytochrome P450 stability.

The Expression Profiles and Deregulation of UDP-Glycosyltransferase (UGT) Genes in Human Cancers and Their Association with Clinical Outcomes

Dong Gui Hu et al.

Summary: The study comprehensively assessed the expression profiles of UGT genes and their impact on patient survival in nearly 30 different cancers primarily derived from non-drug-metabolizing organs. The results showed widespread expression of UGT genes in cancers, indicating the potential of specific UGT genes to serve as prognostic biomarkers or therapeutic targets in cancers.

Epigenetics and microRNAs in UGT1As

Cui-Lan Meng et al.

Summary: UDP-glucuronosyltransferases are important drug-metabolizing enzymes in the human body, with UGT1A family being crucial in glucuronidation reactions. Differences in the distribution and expression of UGT1As among individuals contribute to disease occurrence and metabolic variations. Additionally, epigenetics and miRNAs play a role in regulating UGT1As and impacting sex expression and in vivo distribution, with early life epigenetic changes affecting gene expression throughout life.
