Fetal therapies and trials for lysosomal storage diseases: a survey of attitudes of parents and patients

Marisa E. Schwab et al.

Summary: This study investigated the attitudes of patients with lysosomal storage diseases (LSDs) and their parents towards fetal clinical trials and therapies. The results showed that more than half of the respondents were willing to participate in a phase I clinical trial for fetal enzyme replacement therapy.

Prenatal Somatic Cell Gene Therapies: Charting a Path Toward Clinical Applications (Proceedings of the CERSI-FDA Meeting)

Summary: We are currently living in a golden age of medicine where prenatal diagnosis, fetal therapy, and gene therapy/editing offer the possibility to repair nearly any genetic defect. Prenatal therapies are used in the second or early third trimester for severe, life-threatening disorders. However, the road to clinical prenatal gene therapy is complex.

The rapidly evolving view of lysosomal storage diseases

Giancarlo Parenti et al.

Summary: Lysosomal storage diseases are metabolic disorders caused by deficiencies in lysosomal function components. Understanding of lysosomal biology has evolved from organelles involved in catabolic pathways to dynamic elements involved in multiple cellular functions and capable of adapting to environmental stimuli. Novel technologies based on high-throughput approaches have greatly contributed to the characterization of lysosomal biology and dysfunction.

Lysosomal storage disorders as an etiology of nonimmune hydrops fetalis: A systematic review

Neel S. Iyer et al.

Summary: This study found that the incidence of lysosomal storage disorders in cases of nonimmune hydrops fetalis was 6.6% overall and 8.2% in idiopathic cases. The most common LSDs identified were mucopolysaccharidosis type VII, galactosialidosis, infantile sialic acid storage disease, Gaucher disease, GM1 gangliosidosis, and sialidosis. Over 40% of these LSDs have potential postnatal treatment options, highlighting the importance of early diagnosis and appropriate management.
