Thermoresponsive Ozone-Enriched Spray Gel for Postsurgical Treatment of Hepatocellular Carcinoma

Summary: A thermoresponsive ozone-enriched spray gel is developed to suppress tumor recurrence in hepatocellular carcinoma. The gel contains a perfluorocarbon nanoemulsion that can directly kill tumor cells by generating reactive oxygen species. It induces both ferroptosis and apoptosis by regulating gene expression and inducing lipid peroxidation, which significantly reduces tumor recurrence in Huh-7 tumors.

Penetrative and Sustained Drug Delivery Using Injectable Hydrogel Nanocomposites for Postsurgical Brain Tumor Treatment

Summary: We present an injectable thermoresponsive hydrogel nanocomposite as a potential solution for postsurgical treatment of glioblastoma multiforme (GBM). The hydrogel nanocomposite acts as a soft, deep intracortical drug reservoir, with drug-loaded micelles targeting residual GBM cells. Alternating magnetic fields generated by water-dispersible ferrimagnetic iron oxide nanocubes facilitate penetrative drug delivery. In an orthotopic mouse GBM model, significant tumor growth suppression and improved survival rates are observed.

Summary: Over the past decade, immunotherapy interventions have revolutionized oncology and autoimmune disease fields. Researchers are developing new platforms for immunotherapy that go beyond intravenous delivery strategies. Recent advances include using chemokines to direct immune cells, alternative therapies, and oncolytic viruses. Hydrogel-based materials show promise for diverse biomedical applications, including large molecule drug delivery. Local hydrogel delivery is a promising approach to increase the efficacy and decrease toxicities of immunotherapies. Challenges remain in the translation of hydrogels to the clinic and manufacturing.

Role of Micelle Size in Cell Transcytosis-Based Tumor Extravasation, Infiltration, and Treatment Efficacy

Summary: Transcytosis-based active transport of cancer nanomedicine has been shown to enhance tumor extravasation and infiltration, but how the nanoproperties, particularly particle size, affect transcytosis is unknown. In this study, we used a transcytosis-inducing polymer, OPDEA, to fabricate stable OPDEA-based micelles of different sizes. The smallest micelles (30 nm) showed the fastest transcytosis and most efficient tumor extravasation and infiltration, leading to enhanced antitumor activity compared to larger micelles (140 nm).

Self-assembling paclitaxel-mediated stimulation of tumor-associated macrophages for postoperative treatment of glioblastoma

Summary: We report on a self-assembling paclitaxel filament hydrogel that stimulates macrophage-mediated immune response for local treatment of recurrent glioblastoma. The paclitaxel filaments elicit an immune-stimulating tumor microenvironment and sensitizes tumor to the aCD47-mediated blockade, promoting tumor cell phagocytosis by macrophages and triggering an antitumor T cell response. As adjuvant therapy after surgery, this aCD47/PF supramolecular hydrogel effectively suppresses primary brain tumor recurrence and prolongs overall survivals.

Ferroptosis-Enhanced Immunotherapy with an Injectable Dextran-Chitosan Hydrogel for the Treatment of Malignant Ascites in Hepatocellular Carcinoma

Summary: This study presents an injectable hydrogel drug delivery system based on chitosan hydrochloride and oxidized dextran (CH-OD) for loading sulfasalazine (SSZ) to effectively treat malignant ascites in advanced hepatocellular carcinoma (HCC). The CH-OD-SSZ hydrogel exhibits enhanced cytotoxicity and immunogenic ferroptosis compared to free SSZ. In preclinical models, the intraperitoneal administration of CH-OD-SSZ hydrogel significantly suppresses tumor progression, repolarizes macrophages, activates dendritic cells, and achieves ascites regression when combined with anti-PD-1 immunotherapy.

However, the Enhanced Permeability and Retention (EPR) effect, which was previously considered as the primary extravasation mechanism, is limited and heterogeneous in human tumors. These nanomedicines can actively extravasate into solid tumors by utilizing the transcellular transport mechanism of tumor endothelial cells.
