SERPING1 Variants and C1-INH Biological Function: A Close Relationship With C1-INH-HAE

Christian Drouet et al.

Summary: Hereditary angioedema with C1 Inhibitor deficiency (C1-INH-HAE) is a genetic disorder caused by variants of the SERPING1 gene. These variants result in the failure to control the kallikrein-kinin system (KKS), leading to inflammation. Many of the SERPING1 variants are considered pathogenic, with common variant types including deletions/duplications, point substitutions, and splice site variants.

Overview of SERPING1 Variations Identified in Hungarian Patients With Hereditary Angioedema

Edina Szabo et al.

Summary: This study identified various mutations in C1-INH-HAE patients by determining the concentrations of C1-INH, C4, C1q, and anti-C1-INH antibodies, as well as functional C1-INH activity. It suggests further methods should be applied to cases with unrevealed genetic background.

A robust multiplexed assay to quantify C1-inhibitor, C1q, and C4 proteins for in vitro diagnosis of hereditary angioedema from dried blood spot

Yongquan Lai et al.

Summary: A novel multiplexed assay was developed to measure C1-INH, C1q, and C4 levels in dried blood spots of HAE patients, offering advantages such as less invasive sampling, minimal blood processing, and simultaneous determination of these proteins. The method was fully validated following regulatory guidelines and showed high correlation with ELISA data measured in plasma, providing a new approach for HAE diagnosis.
