Journey to the centre of the lung. The perspective of a mineralogist on the carcinogenic effects of mineral fibres in the lungs

Alessandro F. Gualtieri

Summary: This work provides a review of the bio-chemical mechanisms and adverse effects caused by the inhalation and transportation of mineral fibres in the lungs. It focuses on three known carcinogenic mineral fibres and their behavior in the respiratory tract and pleural cavity. The article discusses the specific crystal-chemical characteristics and properties of these fibres, and critically examines their biological and pathological effects related to lung cancer and malignant mesothelioma. It also highlights open issues, data gaps, and conflicts in the literature and suggests future research directions for a comprehensive model of the carcinogenicity of mineral fibres.

Cytotoxicity of fibrous antigorite from New Caledonia

Elena Gazzano et al.

Summary: Exposure to asbestos and asbestos-like minerals is associated with severe lung diseases. The study investigated the toxicity and cellular effects of fibrous antigorites on murine macrophages and alveolar epithelial cells. The findings revealed significant cellular toxicity of fibrous antigorites, which correlated with their fibrous habit and surface reactivity.

Acute cytotoxicity of mineral fibres observed by time-lapse video microscopy

Dario Di Giuseppe et al.

Summary: This study compared the early cytotoxicity mechanisms of three carcinogenic mineral fibres (chrysotile, crocidolite, and fibrous erionite) using time-lapse video microscopy and in vitro assays. The results showed that fibrous erionite exhibited the earliest toxic effects, while the toxicity of chrysotile and crocidolite increased gradually over time. Chrysotile and crocidolite mainly induced cell death through apoptosis, while erionite exposure may also be associated with a necrotic-like effect.

Characterisation of potentially toxic natural fibrous zeolites by means of electron paramagnetic resonance spectroscopy and morphological-mineralogical studies

Matteo Giordani et al.

Summary: This study explores the characteristics and interaction capabilities of carcinogenic erionite and other zeolite minerals at the liquid/solid interface. It finds that erionite has a high Si/Al ratio and specific surface area, and interacts well with spin probes. Mesolite and thomsonite have lower interaction capabilities and less homogeneous surfaces. Mesolite may pose a potential health hazard to humans, but to a lesser degree compared to erionite. Thomsonite interacts with the biological environment with a low intensity.

Fibrous Ferrierite from Northern Italy: Mineralogical Characterization, Surface Properties, and Assessment of Potential Toxicity

Summary: This study investigates the morphology, crystal structure, chemistry, and surface activity of fibrous ferrierite found in northern Italy using various analytical techniques. The results show that these fibers are breathable, able to reach the alveolar space, and have a high ability to adsorb and interact with chemicals. Fibrous ferrierite can be considered a potential health hazard, requiring precautionary measures.

Summary: This study aimed to investigate the toxicity effects of selected fibrous zeolites on two different cellular models and found cytotoxic effects of erionite in both models. Additionally, the study revealed different toxic behaviors of mesolite and thomsonite fibers, suggesting other potential mechanisms of action.

Summary: This study compares the inflammatory response induced by different carcinogenic mineral fibers in three main macrophage phenotypes. The results reveal that different fibers exert toxic effects through different mechanisms. Furthermore, similar behavior in the production of pro-inflammatory mediators was observed among the three macrophage phenotypes.
