PacC mediates spatial regulation of the phospholipid metabolism in the apple fruit-Penicillium expansum interaction

pH regulates the pathogenicity and extracellular enzyme activity of Gilbertella persicaria on pitaya fruit

Summary: This study investigated the pH change, organic acid secretion, pathogenicity of Gilbertella persicaria on pitaya fruit, and the activities of pectinase and cellulase. The results showed that G. persicaria colonization led to local acidification of pitaya fruit tissue, with pH decreasing from 4.76 to 4.41. Oxalic acid was the main organic acid secreted by G. persicaria. pH 4.4 enhanced the activities of pectinase and cellulase in pitaya fruit. The findings suggest that ambient pH regulates the pathogenicity of G. persicaria, providing potential methods to control fruit decay caused by this fungus.

Small GTPases RasA and RasB regulate development, patulin production, and virulence of Penicillium expansum

Summary: RasA and RasB genes were knocked out in P. expansum, resulting in impaired growth, incomplete spore development, and malformed mycelium. Deletion of PeRasA and PeRasB also led to a significant reduction in patulin content and down-regulation of genes involved in patulin biosynthesis. Furthermore, Delta PeRasA and Delta PeRasB exhibited attenuated virulence on apple and pear fruit and lower gene expression of cell wall degrading enzymes. In conclusion, PeRasA and PeRasB control the MAPK and cAMP signaling pathways to regulate the development, patulin synthesis, and virulence of P. expansum.

Extensive regulation of pH-responsive transcription factor PacC on secondary metabolism contributes to development and virulence of Botrytis cinerea

Summary: This study found that the deletion of the crucial gene BcpacC in Botrytis cinerea strain B05.10 resulted in pH-dependent reduction in mycelial growth, sporulation, and virulence in apple and tomato fruit, and romaine lettuce leaves. Proteomic analysis identified 70 differential proteins involved in various biological processes, including 18 proteins related to secondary metabolism. Gene expression analysis revealed that 37 out of 42 secondary metabolism key enzyme encoding genes were regulated by BcPacC. Additionally, three PacC-regulated polyketide synthase encoding genes were found to contribute to sporulation and virulence in B. cinerea. The extensive regulation of BcPacC on secondary metabolism plays crucial roles in the development and pathogenicity of B. cinerea during adaption to ambient pH.

Preharvest chitooligosaccharide spray alleviates chilling injury in harvested muskmelon fruit by regulating membrane lipid metabolism and activating antioxidant enzyme activity

Summary: This study found that preharvest chitooligosaccharide (COS) spray can alleviate chilling injury (CI) in muskmelon fruit by regulating membrane lipid metabolism and increasing antioxidant enzyme activity. The COS spray up-regulated the expression of CmPLA(2), promoting the release of unsaturated fatty acids and increasing the double bond index (DBI) value. It also down-regulated the expression of CmPLC and CmPLD, increased the levels of phosphatidylcholine (PC) and phosphatidylinositol (PI), and decreased the levels of phosphatidic acid (PA). Furthermore, the COS spray increased the activity of superoxide dismutase (SOD) and catalase (CAT), decreased O-2(-) and H2O2 levels, maintained cell membrane permeability, and reduced MDA levels in fruit during chilling, thereby alleviating CI.

PacC mediated ammonia synthesis contributes to the pathogenicity of Trichothecium roseum by regulating reactive oxygen species and extracellular enzymes

Summary: The Pal signaling pathway is crucial for the adaptation of pathogenic fungi to pH fluctuations and involves several components including PacC. This study investigated the role of TrPacC in Trichothecium roseum, revealing its involvement in pH signal perception, ammonia synthesis, and ROS production. Deletion of TrPacC downregulated the expression of genes related to PacC activation and ammonia synthesis, leading to reduced ammonia accumulation and alkalization ability. Furthermore, TrPacC deletion reduced ROS levels and inhibited the expression of extracellular enzyme genes, resulting in reduced pathogenicity of T. roseum on muskmelon and apple fruit.
