Tracking early lung cancer metastatic dissemination in TRACERx using ctDNA

Christopher Abbosh et al.

Summary: Circulating tumour DNA (ctDNA) can be used to detect and profile residual tumour cells after surgery. A study involving a large number of early-stage non-small-cell lung cancer (NSCLC) patients found that preoperative absence of ctDNA detection is associated with better clinical outcome. Postoperative plasma analysis revealed ctDNA detection in 25% of patients within 120 days after surgery, and 49% of patients who experienced clinical relapse were identified.

A Leaking-Proof Theranostic Nanoplatform for Tumor-Targeted and Dual-Modality Imaging-Guided Photodynamic Therapy

Duo Jin et al.

Summary: This study designed a protein-based leaking-proof theranostic nanoplatform for dual-modality imaging-guided tumor photodynamic therapy. By conjugating chlorin e6 (Ce6) to ferrimagnetic ferritin (MFtn), a biomineralized ferrimagnetic core was generated in the cavity of ferritin, resulting in the formation of a fluorescent ferrimagnetic ferritin nanoplatform (MFtn-Ce6). The distribution and tumor targeting of MFtn-Ce6 can be detected by magnetic resonance imaging and fluorescence imaging. The results showed that MFtn-Ce6 exhibited effective dual-modality imaging and enhanced tumor accumulation and retention of photosensitizer, providing precise tracking and optimal treatment time window for PDT. The co-localization of ferritin and the iron oxide core confirmed the high stability of the nanoplatform in vivo. Mice treated with MFtn-Ce6 demonstrated significant tumor-suppressive activity after laser irradiation.

Temperature non-uniformity detection on dPCR chips and temperature sensor calibration

Martina Ganova et al.

Summary: In this study, a non-contact method for measuring the temperature distribution in a microfluidic-based dPCR chip was proposed. By using a PCR solution with a known melting temperature as a temperature sensor, the temperature distribution was characterized. Compared to other non-contact temperature measurement methods, this method can determine the temperature distribution within the chip and is not affected by the photobleaching effect of fluorescein-based temperature measurement method.

Amplification Efficiency and Template Accessibility as Distinct Causes of Rain in Digital PCR: Monte Carlo Modeling and Experimental Validation

Pieter Berden et al.

Summary: We developed an analytical model that explains the relationship between amplification efficiency, template accessibility, and rain in dPCR assays. Through Monte Carlo simulations, we found that reduced amplification efficiency leads to broader threshold cycle distributions, while template inaccessibility results in an exponential tail rain pattern.

Tracing Tumor-Derived Exosomal PD-L1 by Dual-Aptamer Activated Proximity-Induced Droplet Digital PCR

Bingqian Lin et al.

Summary: The TRACER system utilizes a dual-target-specific aptamer recognition activated in situ connection system combined with ddPCR technology to quantitatively detect tumor-derived exosomal PD-L1 in a wash-free manner, showing significant sensitivity and selectivity. This method has the potential to serve as a more reliable tumor diagnostic marker than total Exo-(PD-L1) and to explore the biological functions of exosomes.
