A novel missense variant in the L2HGDH gene in a cat with L-2-hydroxyglutaric aciduria and multicystic cerebral lesions

Matthias Christen et al.

Summary: A 9-month-old intact male domestic shorthair cat with increasing frequency of generalized tonic-clonic seizures was evaluated. MRI revealed multifocal, small, rounded intra-axial lesions in the subcortical white matter containing fluid similar to cerebrospinal fluid. Whole genome sequencing identified a pathogenic variant in the L2HGDH gene, and increased excretion of 2-hydroxyglutaric acid was detected in urine organic acids. Despite treatment with levetiracetam, the cat died after a seizure 10 days later. This report contributes to the understanding of L-2-hydroxyglutaric aciduria in cats.

SOAT1 missense variant in two cats with sebaceous gland dysplasia

Sarah Kiener et al.

Summary: This study investigated two 4-month-old sibling domestic short haired kittens with various skin abnormalities. Whole genome sequencing revealed a homozygous missense variant in the SOAT1 gene, which has been associated with hair and sebaceous gland defects in mice. This finding suggests that SOAT1 variants may also contribute to hereditary sebaceous gland dysplasias in humans and domestic animals.

Beta-Mannosidosis Is a Cause of Hypomyelination

Deborah L. Renaud

Summary: Beta-mannosidosis is a rare autosomal recessive disorder caused by a deficiency of lysosomal beta-mannosidase due to mutations in the MANBA gene. There is significant clinical variability among the limited number of reported cases. The magnetic resonance imaging (MRI) findings in beta-mannosidosis patients have not been well-documented.

Methemoglobinemia, Increased Deformability and Reduced Membrane Stability of Red Blood Cells in a Cat with a CYB5R3 Splice Defect

Sophia Jenni et al.

Summary: This study describes the clinical, pathophysiological, and molecular genetic features of a cat with hereditary methemoglobinemia. Genetic analyses revealed a single homozygous base exchange in the CYB5R3 gene, resulting in a splice defect and expression of two mutant CYB5R3 transcripts. The affected cat showed characteristics such as increased red blood cell fragility and deformability, glutathione overload, and morphological alterations typical for stress erythropoiesis associated with methemoglobinemia.

A Homozygous MAN2B1 Missense Mutation in a Doberman Pinscher Dog with Neurodegeneration, Cytoplasmic Vacuoles, Autofluorescent Storage Granules, and an α-Mannosidase Deficiency

Garrett Bullock et al.

Summary: This article reports a case of a 7-month-old Doberman Pinscher dog with neurological symptoms and brain atrophy. Pathological examination revealed massive accumulations of vacuolar inclusions in cells throughout the central nervous system, confirming a diagnosis of alpha-mannosidosis. Additionally, the dog exhibited complex ultrastructural features of autophagolysosomes, as well as glial activation indicative of neuroinflammation.

Lysosomal storage disease associated with a CNP sequence variant in Dalmatian dogs

Garrett Bullock et al.

Summary: A progressive neurological disorder characterized by anxiety, pacing and circling, hypersensitivity, cognitive decline, and sleep disturbance has been identified in purebred Dalmatian dogs. The disorder is caused by a loss-of-function mutation in CNP and shares similarities with neuronal ceroid lipofuscinoses.

X-linked myotubular myopathy associated with an MTM1 variant in a Maine coon cat

Matthew A. Kopke et al.

Summary: This study describes the clinical course and genetic findings of a Maine coon cat with X-linked myotubular myopathy caused by an MTM1 missense mutation. Breeders of Maine coon cats should consider screening for this mutation to prevent the production of affected cats and eliminate the variant from the breeding population.

A novel mutation of the CLCN1 gene in a cat with myotonia congenita: Diagnosis and treatment

Christian Woelfel et al.

Summary: This is a case of a 10-month-old castrated male cat with episodic limb rigidity, hypertrophied musculature, and other symptoms. Electromyography and whole genome sequencing identified a mutation in the CLCN1 gene causing myotonic discharges. Treatment with phenytoin resulted in long-term improvement.

Mining the 99 Lives Cat Genome Sequencing Consortium database implicates genes and variants for the Ticked locus in domestic cats (Felis catus)

L. A. Lyons et al.

Summary: Tabby patterns in cats are influenced by at least three loci, including Tabby and Ticked loci. The Ticked locus on cat chromosome B1 is suggested to control the presence or absence of ticked patterns. Variant analysis identified candidate genes and variants associated with the Ticked locus, including Dickkopf Wnt Signaling Pathway Inhibitor 4 (DKK4) variants.

More Than a Moggy; A Population Genetics Analysis of the United Kingdom's Non-Pedigree Cats

Jennifer Irving McGrath et al.

Summary: The study found that non-pedigree domestic cats in the UK, including DSH, DLH, and DSLH cats, are random-bred rather than crossbred or pedigree-admixed. While most of these cats have a lower proportion of HBD segments in their genome compared to pedigree cats, indicating older segments, there is also evidence of recent inbreeding events in a portion of the random-bred cat genomes, suggesting they are not completely immune to inbreeding and its associated health risks.

A domestic cat whole exome sequencing resource for trait discovery

Alana R. Rodney et al.

Summary: The study demonstrates the effectiveness of whole exome sequencing (WES) as a method to investigate genetic disease variants in domestic cats, enabling the identification of pathogenic gene variants for diseases and traits. Utilizing an exome capture design based on the annotated Felis_catus_9.0 genome assembly, WES was performed on 41 cats, revealing both known pathogenic variants and new gene candidate variants.
