Immunohistochemical detection of EGFRvIII in glioblastoma - Anti-EGFRvIII antibody validation for diagnostic and CAR-T purposes

Increased EGFRvIII Epitope Accessibility after Tyrosine Kinase Inhibitor Treatment of Glioblastoma Cells Creates More Opportunities for Immunotherapy

Summary: The number of glioblastoma cases is increasing, but current therapies are ineffective. This study shows that the use of L8A4 antibody with tyrosine kinase inhibitors does not affect its interaction with EGFRvIII and can increase epitope display. The extracellular part of EGFRvIII has plasticity in forming disulfide bridges within monomers and dimers, allowing the L8A4 antibody to recognize both forms. Immunotherapy using L8A4 antibody, combined with TKIs, can potentially improve anti-GB therapy success.

The current landscape of CAR T-cell therapy for solid tumors: Mechanisms, research progress, challenges, and counterstrategies

Summary: The success of CAR T-cell therapy in hematologic cancers has generated enthusiasm for its potential in treating various human cancers. However, there are challenges in using CAR T-cells against solid tumors, such as limited tumor infiltration, unfavorable tumor microenvironments, antigen escape, CAR T-cell exhaustion, and toxic side effects. To overcome these limitations, researchers are developing new CAR designs and integrating multiple therapeutic strategies. This study reviewed the antigens targeted by CAR T-cells and discussed recent advances in engineering and combination therapies to improve their efficacy in solid tumors.
