Role of RNA modifications in carcinogenesis and carcinogen damage response

Michelle Verghese et al.

Summary: Epitranscriptomics studies post-transcriptional RNA modifications and their regulatory enzymes. Among these modifications, N-6-methyladenosine (m(6)A) is the most common internal modification in messenger RNA (mRNA). Advances in technology have renewed interest in this field and led to extensive research on the role of RNA modifications, especially m(6)A methylation, in cancer. Evidence suggests that carcinogens can modulate RNA modifications and dysregulation of the enzymes involved in RNA modifications contribute to carcinogenesis, making them potential drug targets.

lncRNA THAP7-AS1, transcriptionally activated by SP1 and post-transcriptionally stabilized by METTL3-mediated m6A modification, exerts oncogenic properties by improving CUL4B entry into the nucleus

Hai-Ting Liu et al.

Summary: This study identified that THAP7-AS1 is highly expressed in gastric cancer, associated with lymph node metastasis and poor prognosis, and promotes gastric cancer progression through pathways involving SP1 and METTL3. These findings suggest that THAP7-AS1 may be a potential molecular target for gastric cancer therapy.

Summary: A genome-wide association meta-analysis of lung cancer among different populations reveals five new susceptibility loci and identifies population-specific effects of common and rare variants. The study also highlights the potential role of genes such as IRF4 and FUBP1 in promoting DNA damage in lung fibroblasts.

Genetic variants in N6-methyladenosine are associated with bladder cancer risk in the Chinese population

Hanting Liu et al.

Summary: The study investigated the association between genetic variants in m(6)A-switches and bladder cancer risk, identifying a significant effect of a specific SNP in the SOD2 gene on reducing bladder cancer risk. It was also found that SOD2 may regulate its expression through an m(6)A-hnRNP C dependent mechanism, playing a crucial role in suppressing bladder cancer cell growth and invasion.

Genetic variants in m6A regulators are associated with gastric cancer risk

Xiaowei Wang et al.

Summary: The study revealed the crucial regulatory role of m(6)A modification genes in gastric cancer (GC) development, with the SNP rs9906944 C > T in IGF2BP1 significantly associated with a decreased risk of GC. Higher expression of IGF2BP1 was linked to poorer overall survival in GC patients.

Identification of genetic variants in m6A modification genes associated with pancreatic cancer risk in the Chinese population

Pingting Ying et al.

Summary: The study revealed that the rs7495 gene is significantly associated with an increased risk of PDAC, with the G allele promoting the expression of hnRNPC and thus the risk of disease. This provides new insights into the critical role of m(6)A modification in tumorigenesis.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Integrative genomic analysis of N6-methyladenosine-single nucleotide polymorphisms (m6A-SNPs) associated with breast cancer

Zixue Xuan et al.

Summary: This study identified m(6)A-SNPs associated with breast cancer and explored their potential functions. The SNPs rs4829 and rs9610915 were found to potentially be associated with breast cancer due to having eQTL signals, altered gene expression, and proximity to m(6)A modification sites. Further research is needed to clarify how genetic variation affects epigenetic modification and its functions in breast cancer pathogenesis.
