Therapy with oncolytic viruses: progress and challenges

Summary: This article summarizes the advantages of oncolytic viruses (OVs) as an emerging class of cancer therapeutics, including selective replication in tumor cells, delivery of multiple eukaryotic transgene payloads, induction of immunogenic cell death and promotion of antitumor immunity, and a tolerable safety profile that largely does not overlap with that of other cancer therapeutics. Although four OVs and one non-oncolytic virus have been approved for cancer treatment globally, only T-VEC has been widely approved. However, a better understanding of the biology and pharmacology of OVs is needed to fully exploit their therapeutic potential in patients with cancer.

Inhibition of Tumor Metastasis by Liquid-Nitrogen-Shocked Tumor Cells with Oncolytic Viruses Infection

Summary: This study describes a virus-concealed tumor-targeting strategy that enables the delivery of oncolytic viruses (OVs) to lung metastasis through systemic administration. The OVs can actively infect, be internalized, and cloak into tumor cells, which are subsequently treated to eliminate their pathogenicity. This Trojan Horse-like vehicle avoids virus neutralization and clearance in the bloodstream and facilitates tumor-targeted delivery with over 110-fold virus enrichment. Additionally, this strategy can function as a tumor vaccine and initiate endogenous adaptive antitumor effects by increasing memory T cells and modulating the tumor immune microenvironment.

Enzyme-triggered deep tumor penetration of a dual-drug nanomedicine enables an enhanced cancer combination therapy

Summary: Emerging nanomedicines can eliminate cancer cells by promoting excessive intracellular reactive oxygen species (ROS) generation. However, tumor heterogeneity and poor penetration of nanomedicines often result in varying levels of ROS production, where low levels of ROS can actually promote tumor cell growth, reducing the effectiveness of these nanomedicines. In this study, a novel amphiphilic and block polymer-dendron conjugate-derived nanomedicine (GFLG-DP/Lap NPs) was developed, incorporating a photosensitizer (Ppa) for ROS therapy and Lapatinib (Lap) for molecular targeted therapy. By combining ROS therapy with Lap, the synergistic effect effectively kills cancer cells, even in deep tumor regions.

Emerging phagocytosis checkpoints in cancer immunotherapy

Summary: Cancer immunotherapy utilizes immune checkpoints-targeted therapy and adoptive transfer of engineered immune cells to mobilize patients' own immune systems against cancer cells, revolutionizing the field of oncology. Phagocytosis checkpoints, including CD47, CD24, MHC-I, PD-L1, STC-1, and GD2, play essential roles in suppressing immune responses and linking innate and adaptive immunity in cancer immunotherapy. Genetic ablation and blockade of these checkpoints have shown promising results in enhancing phagocytosis and reducing tumor size. CD47 is the most extensively studied phagocytosis checkpoint and has emerged as a promising target for cancer treatment. However, challenges such as anemia and thrombocytopenia due to CD47's ubiquitous expression on erythrocytes need to be addressed. This review discusses the mechanisms, functions, clinical progress, challenges, and potential solutions of phagocytosis checkpoints in cancer immunotherapy, aiming to facilitate combined immunotherapeutic strategies involving both innate and adaptive immune responses.

A physical wiring diagram for the human immune system

Summary: In this study, we systematically mapped the direct protein interactions across a recombinant library that includes most of the surface proteins on human leukocytes, providing a high-confidence and quantitative view of the receptor wiring that connects human immune cells. By integrating our interactome with expression data, we identified trends in the dynamics of immune interactions and constructed a mathematical model that predicts cellular connectivity. We also developed an interactive multi-tissue single-cell atlas to infer immune interactions throughout the body and linked our receptor interactions to functional roles through protein stimulation and microscopy.

In Vivo Tracking for Oncolytic Adenovirus Interactions with Liver Cells

Summary: Hepatotoxicity is a problem in adenovirus cancer therapy, caused by Kupffer cell death and hepatocyte transduction. The interaction between adenovirus and liver cells is not well understood. This study used intravital microscopy to track the infection and immune response in mouse livers after adenovirus injection, and found that Ad5-RGD and Ad5/3 caused macrophage deformation and virus release, but neutrophils and CD8+ T cells did not affect the rate or dynamics of liver infection. Ad5-RGD failed to complete the replicative cycle in hepatocytes, but enhanced liver transduction during hepatic regeneration.
