Toward the next generation of vascularized human neural organoids

Minghui Li et al.

Summary: Thanks to advancements in 3D culture technologies, organoids derived from human stem cells have provided valuable insights into human CNS development and diseases. However, current organoid cultivation protocols still struggle to recapitulate the complexity and maturation of in vivo neural tissues. Recent studies have explored bioengineering techniques such as genetic engineering, coculture, multidifferentiation, microfluidics, 3D bioprinting, and transplantation to enhance organoid maturation and functionality. These methods hold promise in generating more reliable in vitro models for studying human CNS.

Cellular and Engineered Organoids for Cardiovascular Models

Dilip Thomas et al.

Summary: An ensemble of in vitro cardiac tissue models has been developed to aid in the understanding of complex cardiovascular disorders using stem cell differentiation approaches. These models allow for customization and the study of diseased states, with recent progress in the development of various cardiovascular models. However, there are limitations and challenges with current cardiac models that need to be addressed.

Establishing a deeper understanding of the osteogenic differentiation of monolayer cultured human pluripotent stem cells using novel and detailed analyses

Ping Zhou et al.

Summary: Differentiation of osteoblast-like cells from human pluripotent stem cells is a hot topic in bone tissue engineering. Improving the in vitro osteogenic differentiation efficiency of hPSCs by supplementing with functional compounds at desired stages and establishing a stepwise induction system in the future is essential. The study showed heterogeneous cell population throughout the 35-day induction period, with cell telomerase activity and cell cycle analyses providing valuable insights into cell type changes and tumorigenicity.

Human heart-forming organoids recapitulate early heart and foregut development

Lika Drakhlis et al.

Summary: This study successfully generated three-dimensional heart-forming organoids that closely resemble early native heart anlagen, containing myocardial layer, endocardial-like cells, septum-transversum-like anlagen, and distinct anterior versus posterior foregut endoderm tissues. These organoids were used to study genetic defects in vitro and demonstrated a phenotype similar to cardiac malformations seen in transgenic mice with the knockout of NKX2.5 gene.

Cardioids reveal self-organizing principles of human cardiogenesis

Pablo Hofbauer et al.

Summary: Organoids derived from human pluripotent stem cells can spontaneously form chamber-like structures resembling the human heart, which can be controlled and manipulated for studying heart diseases and regeneration. The establishment of self-organizing cardioids provides a powerful platform for mechanistic studies on developmental defects and potential translational research in the future.
